Vliv vazby sterolu na produkci PR proteinů u elicitinu kryptogeinu

Influence of sterol binding to PR proteins production in elicitin cryptogein

Abstract:

¨Kryptogein je 10 kDa elicitor, produkovaný oomycetou Phythopthora cryptogea, který je rozpoznáván specifickým vazným místem na membráně buněk tabáku (Nicotiana tabacum) a spouští obrannou reakci, která zahrnuje produkci aktivních forem kyslíku, změny pH či produkci mikrobicidních látek (fytoalexinů) a expresi PR proteinů. V rámci struktury kryptogeinu se nachází hydrofóbní kavita, která umožňuje vazbu lipidních látkek, jejichž vazba má vliv jednak na strukturu omega smyčky v rámci proteinu a dále na samotnou biologickou aktivitu proteinu. Pomocí molekulového modelování byly vytvořeny mutantní formy kryptogeinu (K13V, V84F, V84FL41F a L41F), které se liší schopností vázat steroly a mastné kyseliny. Cílem této bakalářské práce bylo porovnat expresi PR1a proteinu po aplikaci jednotlivých mutantních forem s expresí u divokého typu kryptogeinu. Pomocí SDS-PAGE a western blottingu byla zjištěna u proteinů L41F a V84FL41F zvýšená aktivace exprese PR1a proteinu 24 hodin po infiltraci v místě aplikace. Na druhou stranu v okolí místa infiltrace byla zjištěna u proteinů K13V, L41F a V84FL41F 48 hodin po aplikaci výrazně nižší akumulace PR1a proteinu. Experiment tudíž potvrdil spojení mezi schopností vázat lipidní látky a schopností exprimovat PR1a protein, kdy však nebyla nalezena přímá spojitost mezi vlivem vazby sterolu a mastné kyseliny.

Abstract:

Cryptogein is 10kDa elicitor, produced by oomycete Phythophtora cryptogea is recognized by specific high afinity binding site on tobacoo plasma membrane and trigger plenty of defence reactions including production of active oxygen species, pH changes or production of antimicrobial substances (phytoalexins) and PR proteins expression. By using molecular modelling were designed mutants of cryptogein (K13V, V84F, V84FL41F a L41F), which differs in their ability to bind sterols and fatty acids. The object of this thesis was to compare the expression of PR1a protein after application of these mutants of cryptogein with protein expression trigger by wild type cryptogein. By using SDS-PAGE and western blotting in proteins L41F and V84FL41F was found higher accumulation of PR1a protein in the site of application 24 hours after infiltration compare to wt cryptogein. On the other hand, 48 hours after infiltration in surroundings of applications site in proteins K13V, L41F a L41FV84F significantly lower accumulation was determined compare to wt cryptogein. Accordingly this experiment confirm relation between lipids binding and ability to trigger PR proteins accumulation, however a direct connection between the sterol and fatty acid binding.
