Mycotoxin adsorbents using carbon nanomaterials

Abstract:

Diplomová práce se zabývá možností použití grafen oxidu jako adsorbentu mykotoxinů. Mykotoxiny jako sekundární metabolity plísní negativně ovlivňují zdravotní stav zvířat a zároveň jejich užitkovost. V teoretické části jsou shrnuty základní poznatky o charakteristice nejvýznamnějších rodů plísní a jejich výskytu v prostředí. Dále jsou charakterizovány mykotoxiny s důrazem na jejich škodlivost vůči hospodářským zvířatům a projevy onemocnění. Práce obsahuje shrnutí obecných doporučení pro eliminaci vzniku mykotoxinů v krmivech jako je dodržování osevních postupů, správné uskladnění a manipulace s krmivy. Hlavní část je věnována představení používaných dekontaminačních materiálů jako jsou jílové minerály, aktivované uhlí a antioxidanty, a také biologické a chemické dekontaminaci krmiv. Důraz je kladen na možnost využití nanomateriálů jako vyvazovačů mykotoxinů, které se dostávají spolu s krmivem do těla zvířat. Jako vhodný materiál se díky svým vlastnostem jeví grafen oxid, který je v posledních letech hodně studován v mnoha oblastech vědy. Praktická část práce se zabývá výzkumem použití grafen oxidu jako vyvazovače mykotoxinů aflatoxinu B1, zearalenonu a deoxynivalenolu, které se velmi často v krmivech vyskytují. Tento výzkum byl realizován v in vitro podmínkách simulace trávení monogastrických zvířat. Výsledky prokazují slibné vyvazovací schopnosti grafen oxidu a jeho budoucí možné použití jako vyvazovače mykotoxinů.

Abstract:

The thesis shows the possibility of using graphene oxide as mycotoxin adsorbent. Mycotoxins as a secondary metabolites of fungi negatively affect the health of animals and their productive performance. The theoretical part of the thesis summarizes the basic knowledge about the characteristics of the most important genera of fungi and their occurrence in environment. Furthermore, mycotoxins are characterized with emphasis on their harmfulness to livestock and are shown disease manifestations. The thesis contains a summary of general recommendations for the elimination of mycotoxins in feed, such as compliance with sowing procedures, appropriate storage and handling of feed. The main part is devoted to the introduction of used decontamination materials such as clay minerals, activated charcoal and antioxidants, as well as biological and chemical decontamination of feedstuffs. Importance is placed on the possibility of using nanomaterials as binders of mycotoxins, because mycotoxins get into the body of animals together with feed. Due to its properties, graphene oxide appears to be a suitable material, which has been widely studied in many areas of science in recent years. The experimental part of the thesis deals with the research of the use of graphene oxide as an adsorbent of the mycotoxins aflatoxin B1, zearalenone and deoxynivalenol, which occur very often in feedstuff. This research was carried out under the in vitro conditions simulating the digestion of monogastric animals. The results demonstrate the promising binding ability of graphene oxide and its possible usability as a mycotoxin binder in future.
