Identification and detection of antibiotic resistance genes in bovine mastitis pathogens in the Czech Republic

Abstract:

Cílem výzkumu bylo vytvoření multiplexních kitů (založených na polymerázové řetězové reakci, PCR) pro detekci genů rezistence k aminoglykosidům (strA, strB), sulfonamidům (sulI, sulII), tetracyklinům (tetA, tetB, tetK, tetM, tetO), makrolidům a linkosamidům (msrA, ermA, ermB, ermC, mefA/E) u významných původců mastitid (Escherichia coli, Staphylococcusaureus, Streptococcus uberis, Streptococcus agalactiae a Streptococcus dysgalactiae). Vyvinuté kity byly aplikovány pro vyhodnocení distribuce genů rezistence k antibiotikům u uvedených druhů izolovaných ze vzorků mléka v České republice.Každý kit se skládal ze sedmi pár primerů. Šest z nich amplifikovalo fragmenty genů rezistence k antibiotikům a jeden pár fragment genu specifického pro příslušný druh. PCR podmínky byly optimalizované pro amplifikaci fragmentů sedmi genů současně v jedné reakci.Ve výzkumu bylo analyzováno 249 vzorků, mezi kterými bylo 111 E. coli, 52 S. aureus a 86 streptokoků. Většina bakterií (60,2 %) nesla alespoň jeden gen rezistenci k antibiotikům a 44,6 % bylo multirezistentních.Vytvořené kity můžou být aplikovány jako diagnostická metoda místo nebo spolu se standardními testy antibiotické rezistenci u uvedených patogenů.

Abstract:

The aim of this research was the development of multiplex polymerase chain reaction (PCR) assays for the detection of aminoglycoside (strA, strB), sulfonamide (sulI, sulII), tetracycline (tetA, tetB, tetK, tetM, tetO), macrolide and lincosamide (msrA, ermA, ermB, ermC, mefA/E) genes of resistance in mastitis pathogens (Escherichia coli, Staphylococcusaureus, Streptococcus uberis, Streptococcus agalactiae and Streptococcus dysgalactiae). One application of the established assays was the investigation of the distribution of antibiotic resistance genes in the above mentioned species isolated from milk samples in the Czech Republic.Each assay consisted of seven primer pairs. Six of them amplified fragments of antibiotic resistance genes and one pair a fragment of a species specific gene. PCR conditions were optimized to amplify seven gene fragments simultaneously in one reaction.In total, 249 isolates were used, among which 111 were positive for E. coli, 52 for S. aureus and 86 for Streptococcus spp. The majority (60.2%) of bacteria carried at least one antibiotic resistance gene and 44.6% were multidrug-resistant.The designed multiplex PCR assays may be applied as diagnostic method to replace or complement standard techniques of antibiotics susceptibility testing in the mentioned pathogens.
