Proteome profiling of silver resistant bacteria Escherichia coli and Staphyloccocus aureus

Abstract:

Hlavní cílem této bakalářské práce bylo studium proteinového profilu vůči stříbrným nanočásticím rezistentních bakterií Escherichia coli a Staphylococcus aureus. Byly zvoleny dva různé přístupy. V prvním byly sledovány intaktní buňky bakterií a jejich proteiny pomocí hmotnostní spektrometrie s ionizací za účasti matrice. Nebyly však nalezeny žádné charakteristické hodnoty m/z, které by umožnily snadné odlišení kmenů vůči stříbru rezistentních a citlivých. U druhého přístupu byla provedena celková analýza proteinů z bakterií za použití extrakce organickými kyselinami, následnou separací proteinů v gelu a jejich identifikací pomocí tandemové hmotnostní spektrometrie. U bakterie E. coli na stříbro rezistentní bylo charakterizováno 53 proteinů, u Staph. aureus 38 proteinů. Vedle flagelinu, hlavního proteinu zodpovědného za rezistenci u E. coli, byly identifikovány další proteiny, které by se mohli podílet na zvýšení životaschopnosti buňky v přítomnosti stříbra. Mechanismus obrany u bakterií Staph. aureus stále není znám, přičemž řada identifikovaných proteinů by se mohla na rezistenci podílet. a to i v souvislosti s jejich strukturou.

Abstract:

The main goal of the bachelor thesis was the study of the protein profile against silver nanoparticles of resistant bacteria Escherichia coli and Staphylococcus aureus. Two different approaches have been chosen. In the first, intact cells of bacteria and their proteins were examined by mass spectrometry with ionization in the presence of the matrix. No characteristic m/z values were found to allow easy differentiation of strains to resistant and sensitive silver. In the second approach, an overall analysis of proteins from bacteria was performed using organic acid extraction, followed by separation of proteins in the gel and their identification by tandem mass spectrometry. A total of 53 proteins were characterized by E. coli on silver-resistant Staph. Aureus 38 proteins. In addition to proteins that might be involved in enhancing cell viability in the presence of silver, other proteins have been identified that could help to increase cell viability. Mechanism of defense for Staph bacteria. Aureus is still unknown, with many identified proteins being able to participate in resistance. And even in relation to their structure.
