Identification of candidate cytoplasmic regulators of Wnt signalling in silico

Abstract:

Náplní bakalářské práce "Identifikace potenciálních cytoplazmatických regulátorů signalizace Wnt in silico" jsou následující úkoly: 1. Studium publikovaných výsledků z "large scale" skrínů siRNA/expresních knihoven, určených k nalezení nových složek dráhy Wnt (Drosophila, Xenopus, myš, člověk) 2. Identifikace savčích homologů potenciálních regulátorů - zejména z proteinových rodin: kináza, fosfatáza, malá GTPáza a GTPáza asociovaný protein (např. GAP a GEF faktory) s využitím bioinformatických nástrojů on line 3. Identifikace nových potenciálních regulátorů signalizace Wnt - tj. proteinů (či jejich homologů), které byly pozitivní ve více, než jednom nezávislém skrínu a nebyly dosud spojovány s přenosem Wnt signálu 4. Charakterizace publikovaných údajů o kandidátních proteinech a navržení hypotéz, jak se nové potencionální komponenty Wnt dráhy podílí na přenosu signálu.

Abstract:

The goal of the bachelor thesis is the following: 1. To sturdy published results of large screens of siRNA/overexpression libraries designed for identification of novel components of the Wnt signalling pathway(fly, Xenopus, mouse, human). 2. To identify mammalian homologues of the potential Wnt pathway regulators - especially with focus on kinases, phosphatases, small GTPases and GAP/GEF factors using bioinformatic tools available online. To identify novel potential reuglators of Wnt signalling - e.g. proteins (or mammalian homologues of the proteins), which showed up in more than one independent screen and were not previously associated with the Wnt signal transduction. 4. To summarize published results about selected candidate regulators and to formulate hypothesis about their possible molecular mode of action in the Wnt signalling.
