Molecular phylogeny of earwigs and application of molecular-phylogenetic approaches in secondary school teaching

Abstract:

Diplomová práce se zabývá infrařádem Protodermaptera, řád Dermaptera (škvoři), jehož zástupci byli v dosud publikovaných fylogenetických studiích zaměřených na škvory zahrnuti jen minimálně. Pro molekulárně-fylogenetickou analýzu byl použit materiál reprezentující všechny čtyři v současnosti rozlišované čeledi infrařádu Protodermaptera (Karschiellidae, Diplatyidae, Haplodiplatyidae a Pygidicranidae). Fylogenetická analýza byla provedena na 5 genech (COI, 18S, 28S, H3 a TUBA) a do analýz bylo zahrnuto celkem 61 druhů (včetně 12 druhů použitých jako outgroup). Ze závěrů vyplývá, že Protodermaptera netvoří monofyletický klád, jelikož do něj spadají také druhy z čeledí Apachyidae, Anisolabididae a Labiduridae, které jsou v současné době řazeny do infrařádu Epidermaptera. Celý tento klád je v sesterské pozici k rodu Allostethella, který je z hlediska platného systému součástí čeledi Labiduridae. Byla zjištěna polyfylie čeledi Labiduridae, která musí být rozdělena na dvě samostatné čeledi odpovídající současným Labidurinae+Nalinae a Allostethinae. Samostatně se vyčlenila skupina sesterská ke zbytku Protodermaptera tvořená rody Esphalmenus a Echinosoma, kterou bude nutno povýšit na úroveň čeledi. Čeleď Karschiellidae se jeví jako sesterská k čeledi Diplatyidae, avšak kvůli omezenému množství dat bude této problematice věnována další studie. Zjištěna byla také monofylie čeledí Diplatyidae+Haplodiplatyidae, která povede k synonymizaci čeledi Haplodiplatyidae. Druhá část práce je věnována zastoupení učiva zaměřeného na využití molekulárních znaků ve fylogenezi organismů ve středoškolských učebnicích biologie. Z výsledků je zřejmé, že se žáci s tímto tématem ve většině učebnic nesetkají a systematika jim je předkládána jako hotová věc. Proto byl vytvořen pracovní list, který žáky seznamuje s důvody a nejnovějšími metodami rekonstrukce systému.

Abstract:

This diploma thesis deals with the infraorder Protodermaptera, the order Dermaptera (earwigs), of which representatives have been included only minimally in previously published phylogenetic studies focused on earwigs. The material used for a molecular phylogenetic analysis representing all four currently differentiated families of the Protodermaptera (Karschiellidae, Diplatyidae, Haplodiplatyidae and Pygidicranidae). The phylogenetic analysis was based on 5 genes (COI, 18S, 28S, H3 and TUBA) with the use of 67 species were used (including 12 species used as outgroups). The conclusion implies that Protodermaptera does not form a monophyletic clade, as it also includes species from the families Apachyidae, Anisolabididae and Labiduridae, which are currently included in the infraorder Epidermaptera. This whole log is in a sister position to the genus Allostethella, which is a part of Labiduridae in terms of the current system. The polyphyly of the family Labiduridae was determined and must be divided into two separate families corresponding to the current Labidurinae+Nalinae and Allostethinae. A group sister to the rest of Protodermaptera clustered separately, consisting of the genera Esphalmenus and Echinosoma, will need to be raised to the family level. Karschiellidae appears to be a sister group to Diplatyidae, but due to limited data, further studies will be devoted to this issue. Moreover, a monophyly of Diplatyidae+Haplodiplatyidae was also found, which will lead to synonymization of Haplodiplatyidae. The second part of the diploma thesis deals with the representation of the school curriculum focused on the use of molecular features in the phylogeny of organisms in high school biology textbooks. It is evident from the results that students usually do not encounter this topic in most of the textbooks and the systematics is presented to them as a done deal. Therefore, a worksheet was created as a part of the thesis that introduces students to the latest methods and the reasons for the reconstruction in the phylogenetic system.
