Function of alternative splicing of the MPK13 gene during flowering in Arabidopsis thaliana

Abstract:

Práca sa venuje štúdiu mitogen-aktivovaných proteín kináz (MAPKs). Dôležitým komponentom rastlinnej signalizácie zodpovedných za prenos signálu a tvorbu odpovedi na podmienky okolia. Konkrétne sa práca zameriava na dve ortológne kinázy prvého rádu AtMPK13 a MsMMK3. V práci boli pripravené vstupné komponenty pre molekulárne klonovanie trojkomponentovou reakciou systému "Multisite Gateway". Jednokomponentovou reakciou bol pripravený jeden fluorescenčne značený konštrukt génu MPK13. Prebehla úspešná validácia daného konštruktu v bode tranzientnej transformácie listov N. bentahmiana. Taktiež boli vykonané analýzy prítomnosti a následne aj hladín jednotlivých MPK13 transkriptov, za účelom overenia dostupných T DNA inzerčných línii na pracovisku a s cieľom vysvetliť pozorované rozdiely v predchádzajúcej bakalárskej práci. Transkripty boli detegované vo všetkých troch mutantných líniách a dané fenotypové analýzy boli následne rozpracované aj v diplomovej práci, pričom poukázali na veľmi mierne fenotypové rozdiely medzi jednotlivými líniami. Okrem práce zameranej na gén MPK13 bola počas diplomovej práce vykonaná validácia potenciálnej CRISPR KO mutantnej línie v ortológnom géne MMK3 Lucerny siatej. V tomto prípade bola identifikovaná jedna mutantná rastlina s prítomnou editáciou v oboch alelách kódujúcich daný gén. Tento rastlinný materiál bol následne zmnožený dvomi rôznymi metódami a uchovaný pre ďalšie potenciálne experimenty.

Abstract:

The thesis is focused on the study of mitogen-activated protein kinases (MAPKs), an important component of plant signaling responsible for signal transduction and the generation of responses to environmental conditions. Specifically, the work focuses on two orthologous first-order kinases, AtMPK13 and MsMMK3. In this work, entry components for molecular cloning were prepared using a three-component reaction of the Multisite Gateway system. A single fluorescently labeled construct of the MPK13 gene was prepared using a one-component reaction. Successful validation of this construct was carried out at the level of transient transformation of N. benthamiana leaves. Analyses of the presence and subsequently also the levels of individual MPK13 transcripts were performed in order to verify the available T-DNA insertion lines at the workplace and to explain the observed differences reported in the previous bachelor's thesis. Transcripts were detected in each of the three mutant lines and phenotypic analyses were further elaborated in the diploma thesis. These analyses revealed only very subtle phenotypic differences among the individual lines. In addition to the work focused on the MPK13 gene, validation of a potential CRISPR knock-out mutant line in the orthologous MMK3 gene of alfalfa was performed during the thesis. In this case, one mutant plant was identified carrying an edit in both alleles encoding the given gene. This plant material was subsequently propagated using two different methods and preserved for further potential experiments.
