Antinutritional factors in pea

Abstract:

Hrách patří mezi základní zemědělské plodiny pěstované především v Evropě, a to pro svůj vysoký obsah proteinů. V semenech hrachu se rovněž nachází i antinutriční faktory, látky, jimž je věnována teoretická část práce. Zahrnuty jsou informace o jejich typech, funkci v semenech a biologických účincích. Dále jsou uvedeny i metody stanovení těchto sloučenin. Experimentální část práce byla věnována osvojení si metody spektrofotometrického stanovení obsahu inhibitoru trypsinu. Zejména byl kladen důraz na optimalizaci metody s využitím spektrofotometrického readeru a také na celkovou minimalizaci experimentu. Zavedenou metodou byl posléze stanoven obsah inhibitoru trypsinu ve vybraných komerčně dostupných druzích luskovin a také v odrůdě sóji poskytnuté VÚRV. Právě zde byl zaznamenán nejvyšší obsah inhibitoru, v hrachu byl naopak jeho obsah nejnižší. Zároveň byla osvojena metoda kvantitativní analýzy kyseliny fytové. Ke stanovení byla využita nejprve kapilární elektroforéza, poté byla nahrazena spektrofotometrií. Vysoký obsah kyseliny fytové byl zaznamenán u druhů, v nichž byl vysoký i obsah inhibitoru trypsinu.

Abstract:

Pea is one of the staple crops. This leguminous crop is predominantly cultivated in Europe. Its significance lies in the high protein content. It also contains special compounds called antinutritional factors. These compounds are the main topic of the theoretical part of this thesis. The theoretical part also provides information about types of antinutritional factors, their function in seeds, and also the biological effects. Furthermore, there are also listed the main methods for determining these compounds. The aim of the experimental part of this thesis was to optimize the method of spectrophotometric determination of trypsin inhibitor content. The optimization was done by minimizing the experiment and also by using a spectrophotometric reader for the determination. The optimized method was then used for the determination of the content of trypsin inhibitor in selected commercially available legume species and the soybean variety provided by VÚRV. The soybean variety contained the highest amount of trypsin inhibitor. On the other hand, the pea contained the lowest amount of trypsin inhibitor. Another aim of the experimental part was to develop the method for the quantitative determination of phytic acid. This was done by using capillary electrophoresis first, then the spectrophotometric determination was used instead. The samples with the high trypsin inhibitor content contained a high amount of phytic acid.
