Synthesis of (1\kur{R}, 2\kur{R})-\kur{N}-benzylcyklohexan-1,2-diamine

Abstract:

Byla provedena literární rešerše týkající se přípravy a charakterizace cyklohexan-1,2-diaminů dále zaměřená na katalytické využití nikelnatých komplexů opticky čistých substituovaných cyklohexan-1,2-diaminů. Byly shrnuty a zhodnoceny známé metody přípravy (1R,2R)-N-benzylcyklohexan-1,2-diaminu. Přímou benzylací a následnou redukcí kyanborohydridem sodným byl připraven racemický trans-N-benzylcyklohexan-1,2-diamin z trans-cyklohexan-1,2-diaminu a (1R,2R)-N-benzylcyklohexan-1,2-diamin z opticky čistého (1R,2R)-cyklohexan-1,2-diaminu. Byla také prověřena metoda přípravy, kde byl výchozí opticky čistý (1R,2R)-cyklohexan-1,2-diamin ochráněn ftaloyl skupinou, benzylován a následně podroben odchránění ftaloyl skupiny. Výsledné produkty byly charakterizovány pomocí NMR spektroskopie, FT-IR spektroskopie nebo hmotnostní spektrometrie ve vysokém rozlišení

Abstract:

The literary research was carried out on the preparation and characterization of cyclohexane-1,2-diamines further aimed at the catalytic application of nickel complexes of optically pure substituted cyclohexane-1,2-diamines. The known methods for the preparation of (1R,2R)-N-benzylcyclohexane-1,2-diamine were summarized and evaluated. Direct benzylation and subsequent reduction by cyanoborohydride were conducted on racemic trans-cyclohexane-1,2-diamine to give trans-N-benzylcyclohexane-1,2-diamine and on optically pure (1R,2R)-cyclohexane-1,2-diamine to give (1R,2R)-N-benzylcyclohexane-1,2-diamine. The method of preparation whereby the starting optically pure diamine was protected by phthaloyl, benzylated and subsequently deprotected was also tested. Final products were characterized by NMR spectroscopy, FT-IR spectroscopy or matrix-assisted laser desorption/ionization.
