Neuroendocrine differentiation of prostate and colon cancer cells

Abstract:

Nádorové onemocnění prostaty a tlustého střeva patří k celosvětově rozšířeným onemocněním. Z hlediska rozvoje nádorového onemocnění jsou důležité procesy ovlivňující mikroprostředí nádoru. Jedním z nich může být i neuroendokrinní diferenciace. Cílem této práce je shrnout dosavadní poznatky o tomto ději, popsat roli neuroendokrinních buněk a látek, které vylučují, na progresi nádoru. Dalším důležitým faktorem pro vznik a rozvoj nádorového onemocnění je změna exprese celé řady genů, jejichž produkty mají různé biologické funkce. Použití technologie microarray umožňuje sledovat komplexní změny genové exprese na úrovni transkripce a porovnat rozdíl v genové expresi mezi zdravou a rakovinnou tkání. Tato metoda generuje velké množství dat, která nebývají zcela vytěžena. Proto vznikají veřejně dostupné databáze, které tato data sdružují, zpřístupňují je dalším vědcům a umožňují jejich re-analýzu. Součástí této práce je re-analýza výsledků genové exprese na základě studií z veřejně dostupné databáze Oncomine u vybraných genů, jejichž exprese je spojena s procesem neuroendokrinní diferenciace. V experimentální části práce byl ustanoven model neuroendokrinní diferenciace buněčné linie LNCaP in vitro, který bude dále umožňovat studium a možnosti ovlivnění tohoto procesu.

Abstract:

Both prostate cancer and colon cancer are world-wide extended diseases. Processes, which occur in the tumor microenvironment, can play an important role in the progression of the disease. The aim of this thesis is to summarize present knowledge about neuroendocrine differentiation and describe the role of factors produced by neuroendocrine cells in the tumor progression. The initiation and progression of tumor can be influenced by the deregulation of expression of broad spectrum of different genes, whose products execute different function. Application of microarray technology is enabling to analyze complex changes in gene expression on the level of transcription and compare gene expression pattern of normal healthy and cancer tissues. However, this method generates a large number of data, which usually are not fully mined. That is the reason why new public databases, which associate data from studies concerning with gene expression originates on the internet. Their goal is to provide the large spectrum of the data to other researchers for data-mining and re-analysis. In this thesis, the public database Oncomine was used for re-analysis of expression data for selected genes associated with neuroendocrine differentiation and correlates them with stage of cancer progression. In the experimental part, the model of neuroendocrine differentiation of LNCaP cells was established. This model will serve in the future for another study of the mechanisms of neuroendocrine differentiation and its modulation.
