Finding of a Mathematical Model for the Description of the Influence which has the Interference of Haemoglobin on the Selected Biochemical Parameters

Abstract:

Práce se zabývá nalezením vhodného modelu vícenásobné regrese, který by popisoval působení mechanismu interference hemoglobinu v séru na stanovení analytu. Vliv interference byl sledován na 12 vybraných analytech z celkového spektra vyšetření. Kvalita regresního modelu byla hodnocena pomocí koeficientu determinance R2 a analýzou rozptylu pomocí Fischer-Snedecorova kritéria F. Výsledky práce ukazují téměř totožný vliv hemolýzy na enzymy alaninaminotransferázu a aspartátaminotransferázu, podobný vliv u hořčíku, cholesterolu a draslíku a zároveň podobný vliv triacylglycerolů a celkové bílkoviny, ale s opačným působením vlivu hemoglobinu. Největší vzájemné působení hemoglobinu a analytu bylo prokázáno u celkového bilirubinu, dále pak u železa a naopak nejnižší u albuminu. Nejméně vhodný model byl nalezen u stanovení sodíku, kde byl nejnižší koeficient determinance i hodnota F-testu. Závěry této studie přispěly k vysvětlení působení interference hemoglobinu na stanovovaný analyt.

Abstract:

This diploma work is focused on founding a suitable model of multiplex regression, which will interpret effect of mechanism interference haemoglobin in the serum for determination an analytical sample. The influence of interference was monitored on the twelve selected analytical samples from the total examination spectrum. Quality of the regression model was valuated by index determinate R2 and by analysis of variance through the Fischer-Snedecor criteria F. Results of this work indicate nearly equal influence hemolysis on enzyme alanineaminotransferase and aspartateaminotransferase, similar effect at magnesium, cholesterol and kalium and at once similar influence triacylglyceride and general protein, but with the reverse influence effect of haemoglobin. The biggest interaction betwen haemoglobin and analytical sample was documented at the total bilirubin, then with ferrum and by contrast lowest at albumin. The least suitable model was found in sodium determination , where was the lowest index of determination and a F-test value too. Results of this study assist explain the effect of interference haemoglobin on determination an analytical sample.
