Analysis of markers for grain hardness in triticale

Abstract:

Tvrdost zrna patří mezi důležité kvalitativní parametry obilovin, které ovlivňují jejich koncové využití i tržní hodnotu. Proto je u tritikale, charakteristického spíše měkčím zrnem, důležité porozumět mechanismům determinujícím tvrdost zrna a aplikovat nové poznatky ve šlechtitelských programech. Za genetickou podstatu tvrdosti zrna u pšenice a příbuzných druhů je považován Ha lokus s trojicí genů Pina, Pinb a Gsp-1. Produkty těchto genů svými vazbami ovlivňují strukturu endospermu, kde se nacházejí. Experimentální část této práce směřovala do 3 hlavní oblastí. U souboru vzorků ozimé formy tritikale z let 2015 a 2016 byla s využitím metody PSI změřena tvrdost zrna a tato měření byla statisticky zhodnocena. Poté byla provedena analýza vybraných molekulárních markerů metodou PCR. Vybrané produkty těchto PCR reakcí byly dále sekvenovány. Analýza těchto DNA sekvencí pomocí volně dostupných bioinformatických nástrojů vedla k objevení významných homologií s již popsanými sekvencemi v databázi Genbank.

Abstract:

Grain hardness is one of the important qualitative traits in cereals that affect their end-use and market price. Therefore in triticale, characteristic with rather softer grain, it is important to understand mechanisms determining grain hardness and apply new findings in breeding programs. Genetic basis of grain hardness in wheat and related species is considered to be Ha locus with three genes Pina, Pinb and Gsp-1. Products of these genes with their bonds affect the structure of endosperm, where they can be found. Experimental part of this thesis aimed at 3 main fields. In set of winter triticale samples from years 2015 and 2016 grain hardness was measured using PSI method and statistically evaluated. Then analysis of selected molecular markers via PCR method was conducted. The analysis of these DNA sequences using available bioinformatic tools lead to finding significant homologies with known sequences in the Genbank database.
