Molecular mechanisms of axon guidanceregulation in development and neuroregeneration

Abstract:

Axonální navádění je vysoce dynamický proces probíhající za účelem nalezení vhodného synaptického partnera, což je základní podmínka pro přežití a integraci neuronu jakožto strukturní a funkční součásti nervové soustavy. Cílem této práce je podrobně popsat molekulární mechanismy, které jsou podkladem tohoto procesu, počínajíc vlivem složek extracelulární matrix a končíc shrnutím dosavadních informací o vybraných faktorech ovlivňujících motilitu růstového konusu in vitro a in vivo. Práce začíná krátkým náčrtem vzniku nervové soustavy obecně a dále je tematicky rozdělena do dvou částí: první část je věnována mechanismům axonálního navádění jako součásti ontogeneze nervové soustavy, v další části pak následuje popis regulace růstu axonů při neuroregeneraci.

Abstract:

Axon guidance is a highly dynamic process occurring for the purpose of finding an appropriate synaptic partner, what is the main condition for survival and integration of neuron as a structural and functional component of the nervous system. The goal of this thesis is to describe molecular mechanisms, which are the basis of this process, beginning with the influence of components of extracellular matrix and ending with conclusions of today’s information on selected factors influencing the growth cone motility in vitro and in vivo. The thesis begins with a short introduction of development of nervous system in general and thereinafter is divided into two parts: the first one is dedicated to the mechanisms of axon guidance as a part of ontogenesis of the nervous system and the next one to the regulation of axon pathfinding in neuroregeneration.
