The role of extracellular L-arginine in phagocyte-mediated inflammatory responses

Abstract:

Cílem této diplomové práce bylo popsání vlivu různých extracelulárních koncentrací L-argininu (0 – 400 μM) jak na nestimulované makrofágy, tak i na makrofágy stimulované pomocí bakteriálního endotoxinu. Pozornost byla věnována zejména působení L-argininu na expresi a aktivaci povrchových receptorů spřažených s G-proteinem, sledováním aktivace G-proteinů, změny intracelulární koncentrace vápníkových iontů a také aktivace fosfolipasy C. V této souvislosti byly dále zkoumány změny v aktivaci intracelulárních signálních drah, které jsou zapojeny do aktivace makrofágů pomocí bakteriálního endotoxinu. Pro tuto práci byly využity myší peritoneálními makrofágy RAW 264.7. Pro studium receptorů byla ovšem důležitá také příprava buněk RAW 264.7 se sníženou expresí obou receptorů (pomocí elektroporace) jako i použití inhibitorů jednotlivých sledovaných drah. Pro experimenty byly využity metody jako Griessova kolorimetrická metoda (stanovení produkce NO), fluores-cenční metoda pro stanovení intracelulární koncentrace vápníku, spektrofotometrické stanovení aktivity fosfolipasy C a metoda Western blot (exprese sledovaných proteinů).

Abstract:

The aim of this thesis was to describe the effect of different L-arginine concentrations (0 – 400 μM) on the physiological functions of resting macrophages and macrophages active-ted by bacterial endotoxin. The attention was focused mainly on the L-arginine-dependent activation of G- protein coupled receptors, which was monitored on the level of G-protein activation, concentration of intracellular calcium and activation of phospholipase C. The activation of intracellular signaling cascades, responsible for macrophage stimulation with bacterial endotoxin, was also examined. RAW 264.7 macrophages, as well as the macrophages with down regulated expression of particular G-protein coupled receptors, were used in this study. Physiological functions and activation of signaling pathways were assessed by different methods. The Griess reaction was used for determination of NO production, intracellular concentration of calcium was monitored using fluorescent probe, activity of phospholipase C was determined by colorimetric method and expression of proteins was assessed using Western blot.
