Characterization of the intestinal microbial flora by culture independent techniques

Abstract:

Střevní mikroflóra je soubor všech mikroorganismů, které se vyskytují v trávicím traktu v rovnováze. V této práci jsem se zabývala studiem střevní mikroflóry nosnic Lohman Braun a cílem bylo zjistit, zda existují rozdíly ve složení střevní mikroflóry vzhledem k věku nosnic. Vzorky trusu byly odebírány ze slepého střeva nosnic starých 12, 34, 51 týdnů a k analýze byla použita na kultivaci nezávislá metoda Terminal Fragment Length Polymorphism, která je založena na amplifikaci konzervativních úseků bakteriálních genů pro 16S rRNA a následném štěpení restrikčními endonukleázami. Z dosažených výsledků je tedy zřejmé, že se střevní mikroflóra v průběhu života nosnic dynamicky mění, protože nosnice z každého analyzovaného období měli jinak zastoupené baktérie charakteristické délky fragmentů a u nosnic starých 12 a 51 týdnů byly detekovány fragmenty typické pouze pro daný věk.

Abstract:

Intestinal microflora consists of a number of microorganisms which originate from intestinal tract and are located there in state of balance. This thesis is dedicated to analysis of intestinal microflora of Lohman Braun hens, and the main purpose was to examine potential differences in composition of intestinal microflora in respect to age of the hens. Samples of feces used for analysis were collected from appendix of the hens aged 12, 34 and 51 weeks, and the method utilized in this process was culture-independent technique termed Terminal Fragment Length Polymorphism which is based upon amplification of fragments of bacterial genes 16S rRNA and followed by fragmentation of restriction enzyme digestion. The results confirm that intestinal microflora is of altering nature in course of the hens’ life span because the hens of different age had different number of fragments in all stages of analysis, and with the hens aged 12 and 51 weeks, specific fragments typical for the respective age were detected.
