Importance of N+K+ATPases in biological processes of animal cells

Abstract:

Iontové pumpy (ATPázy) jsou proteinové komplexy včleněné do buněčných membrán, přenášející ionty z extracelulárního do intracelulárního prostředí a naopak proti jejich koncentračnímu gradientu. Takový přenos vyžaduje energii ve formě ATP. Nejvíc rozšířeným druhem iontových pump je sodno-draselná (Na/K-ATPáza). Hlavní rolí tohoto enzymu je udržení nízké koncentrace iontů Na+ a vysoké koncentrace iontů K+ uvnitř buňky. Tato práce se zaměřuje na popis mechanismu, struktury a funkce Na/K-ATPáz v živočišném organismu a skládá se z několika kapitol. První část se zabývá principem Na/K-ATPázy a studiem mechanismu přenosu iontů přes membránu. Druhá část popisuje strukturu studované pumpy, její podjednotek a rozdělení na izoformy. Další kapitola je zaměřena na výskyt a role Na/K-ATPázy a její izoforem v jednotlivých tkáních živočišného organismu. Dále je popsán význam ATPáz v termogenezi. Poslední kapitola zahrnuje návrh experimentu, ve kterém by se sledovala exprese Na/K-ATPázy ve vztahu k tepelné odolnosti skotu.

Abstract:

Ion pumps (ATPases) are protein complexes incorporated into cell membranes, which transport ions from the extracellular to the intracellular environment and vice versa against their concentration gradient. This transmission requires energy in the form of ATP. The most common type of ion pumps is sodium-potassium (Na/K-ATPase). The main role of this enzyme is to keep a low concentration of Na+ ions and a high concentration of K+ ions inside the cell. This work focuses on the description of the mechanism, structure and function of Na/K-ATPase in the animal organism and consists of several chapters. The first part deals with the principle of Na / K-ATPase and the study of the mechanism of ion transfer across the membrane. The second part describes the structure of the studied pump, its subunits and the division into isoforms. The next chapter focuses on the occurrence and role of Na / K-ATPase and its isoforms in individual tissues of the animal organism. The last chapter describes the importance of ATPases in thermoregulatory processes. The last chapter includes the design of an experiment in which would be monitored the expression of Na/K-ATPase in relation to the heat resistance of dairy cattle.
