Translocation of complex peptides across phospholipid membranes

Abstract:

Efektivní přenos specifických molekul přes buněčnou plazmatickou membránu je nezbytný pro životaschopnost buněk a pro doručování léčiv. Nicméně není stále zcela jasné, jaké molekulární vlastnosti jsou nutné pro spontánní translokaci a jestli by bylo možné transportovat molekuly, které tyto vlastnosti nemají. V této práci se zaměřujeme na translokaci komplexních peptidů přes fosfolipidové membrány pomocí počítačových simulací využivajícíh zhrubené modely a zkoumáme faktory ovlivňující účinnost a mechanismus translokace. Ukazujeme, že strategické začlenění hydrofobních vlastností do peptidů může zlepšit jejich schopnost translokace a jaký je pak nejvýhodnější mechanismus translokace. Celkově tato práce poskytuje podrobný vhled do translokace komplexních peptidů přes fosfolipidové membrány, odhaluje jejich výhodné vlastnosti pro samovolnou translokaci. Tyto poznatky mohou být využity při návrhu peptidových terapeutik a přispívají k našemu porozumění základním mechanismům biomolekuláního transportu.

Abstract:

Efficient translocation of specific molecules across cell the plasma membrane is crucial for cell viability and drug delivery. However, it remains unclear what molecular properties are required for spontaneous translocation and whether it would be possible to transport molecules that do not have these properties. In this study, we investigate the translocation of complex amphiphilic peptides through phospholipid membranes using computer simulations with coarse-grained models and investigate the properties affecting their efficiency and mechanism of translocation. We show that strategic incorporation of hydrophobic moieties into peptides can improve their translocation ability and then what is the most favorable translocation mechanism. In summary, this study provides a detailed insight into the translocation of complex peptides across phospholipid membranes, revealing advantageous properties for spontaneous translocation. These findings could be utilized in the design of peptide therapeutics and contribute to our understanding of the underlying mechanisms of biomolecular transport.
