Arenaviruses and hantaviruses of African small mammals

Abstract:

Zástupci rodu Arenavirus a Hantavirus jsou jednořetězcové RNA viry, které u člověka vyvolávají onemocnění jako je krvácivá horečka či hantavirový plicní syndrom, čímž představují významné epidemiologické riziko. K přenosu těchto virů dochází převážně při kontaktu s exkrety či sekrety jejich přirozených hostitelů, hlodavců nebo hmyzožravců. Výjimku tvoří arenavirus Tacaribe a dva nově objevené hantaviry z Afriky, izolované z letounů. Zatímco arenaviry jsou intenzivně studovány v Americe a hantaviry v Evropě či Asii, Afrika byla v posledních desetiletích značně opomíjena. Současné studie však ukazují, že diverzita těchto virů je stále velmi málo známá a je potřeba jí věnovat patřičnou pozornost. V práci jsou sumarizovány dosavadní poznatky o arenavirech a hantavirech, se zvláštním důrazem na viry Starého světa. Dalším z cílů této práce bylo optimalizovat protokol pro genotypizaci virů z krevních vzorků afrických drobných savců za účelem identifikace nových virových druhů či linií. Molekulárně genetické analýzy prvních 201 vzorků z Etiopie odhalily výskyt dvou nových linií jednoho arenaviru a zcela nového druhu hantaviru. Tyto výsledky jsou velmi perspektivní a budou sloužit mj. k pochopení evoluce těchto významných skupin virů.

Abstract:

Members of genera Arenavirus and Hantavirus are single-stranded RNA viruses that can cause diseases such as hemorrhagic fever and cardiopulmonary syndrome in humans, and thus represent a significant epidemiological risk. These viruses are mainly transmitted through contact with secreta or excreta of their natural hosts, rodents or soricomorphs. The exceptions are the Tacaribe arenavirus and two novel African hantaviruses, isolated from bats. While arenaviruses are intensively studied in Americas and hantaviruses in Europe and Asia, Africa has been largely overlooked in last decades. Recent studies indicate that the diversity of these viruses is still much underestimated and attention should be given to their description. This thesis summarizes the current knowledge about arenaviruses and hantaviruses, with special emphasis on Old World viruses. Another objective of this study was to optimize the protocol for genotyping viruses from blood samples of African small mammals for the identification of novel viral strains or species. Molecular screening of the first 201 samples from Ethiopia revealed the presence of two novel strains of arenavirus and the novel hantavirus species. These results are very promising and will be important for understanding the evolution of these important groups of viruses.
