Abstract:

Technologie microarrays neustále ukazuje, že v době post genomových technologií bude hrát jednu z hlavních rolí. Je prakticky nezastupitelná ve výzkumu včasné diagnostiky některých velmi vážných a nebezpečných chorob (rakovina, leukémie atp.). Obrazová analýza je klíčovou částí analýzy dat pořízených technologií microarrays. Přesto v této oblasti, především díky její krátké existenci, zatím neexistuje žádný komplexní a robustní přístup. V tezích mé disertační práce jsou publikovány nové přístupy obrazové analýzy. Důraz je kladen na vzájemnou kompatibilitu jednotlivých fází obrazové analýzy (filtrace, adresace, segmentace, kontrola kvality a kvantifikace). Celkový přístup lze pak jednoduše složit z metod v práci uvedených a na základě jejich popisu. Nejrozsáhlejší pasáž je věnována adresaci, neboť v ní není připustitelná ani nejmenší odchylka. V práci je prezentován nový algoritmus, který je aplikovatelný na obecné microarrays sklíčko. I on je popsán modulárně a jednotlivé jeho části je možné použít k vylepšení již existujících algoritmů. Celkově je možné v této práci nalézt kompletní metodiku potřebnou k analýze obrazů pořízených technologií microarrays včetně popisu návazností na další fáze analýzy dat microarrays. Díky tomu je přístup sestavení s pomocí této práce přímo použitelný v reálném výzkumu založeném na microarrays.

Abstract:

Microarrays is a very promising technology of the post-genome era and will play one of the most important roles especially within the research of early diagnostics of dangerous and serious diseases (cancer, leukemia etc.). Image analysis is a crucial phase of microarrays data processing. There is a need of robust approach. This area is still very young and therefore the complex solution is still missing. There are many novel approaches published in this work. Their compatibility was one of the goals and therefore the overall approach (consisting of filtering, addressation, segmentation, quality control and quantization) can be assembled using the described methods. The most of the thesis is dealing with addressation. This phase doesn’t tolerate any imprecision. The novel approach is presented in the thesis which is applicable to generic microarray slide. Its description is modular and the interim phases can be used separately in order to improve the existing approaches. This thesis presents complete microarrays image analysis methodic. All methods are described with having the following parts of microarrays data analysis in mind and therefore the approach assembled using this thesis can be applied in real research based on microarrays.
