Structure - function studies of lectins from opportunistic bacteria

Abstract:

Lektiny jsou proteiny neimunitního původu, které specificky rozpoznávají cukerné struktury. Některé rostlinné a živočišné patogeny využívají těchto protein-cukerných interakcí k jejich strategii při invazi do hostitele. Předkládaná disertační práce přibližuje studium mechanismů molekulárních interakcí mezi lektiny produkované bakteriálními patogeny Burkholderia cenocepacia a Ralstonia solanacearum s jejich ligandy. Metody jakožto krystalografie či resonance povrchového plasmonu, titrační mikrokalorimetrie a další byly použity k charakterizaci dvou studovaných proteinů. Ralstonia solanacearum je rostlinný bakteriální patogen, který produkuje tři rozpustné lektiny, nazvané RSL, RS-IIL a RS20L. Všechny tři byly již dříve krystalovány, ale charakterizace posledního z nich, RS20L, nebyla ještě dokončena. Během studia byla optimalizována produkce tohoto lektinu a jeho fyzikálně-chemické vlastnosti byly analyzovány pomocí 2D SDS-PAGE / MALDI-MS a diferenciální skenovací kalorimetrie. Vzhledem k tomu, že řada uskutečněných experimentů vedoucí k určení specifity a afinity lektinu nebyla úspěšná, je třeba navrhnout alternativní strategii. Druhá a hlavní část disertační práce je věnována lektinu z lidského podmíněného patogenu Burkholderia cenocepacia zodpovědného za vysokou úmrtnost u pacientů trpících cystickou fybrózou a chronickým granulomatózním onemocněním. Lektin BC2L-C je 28 kDa protein složený ze dvou oddělených domén, které byly zvlášť klonovány a exprimovány v E. coli a následně charakterizovány. C-terminální doména vykazuje vysokou sekvenční a strukturní podobnost s lektinem PA-IIL z bakterie Pseudomonas aeruginosa a rozpoznává s vysokou afinitou D-manosylované sacharidy. Na druhou stranu N-terminální doména je překvapivě také schopna vázat cukry, avšak s vysokou specifitou k L-fukosylovaným oligosacharidům, jako jsou například antigeny krevních skupin, typ H nebo Lewis. Vyřešená krystalová struktura N-terminální domény v komplexu se seleno-methy-fukosidem představuje trimerní seskupení, které zatím nebylo u lektinů pozorováno, ale které je vysoce podobné s TNF- nebo C1q složkou komplementu. Díky zmíněným neobvyklým vlastnostem může být lektin BC2L-C klasifikován jako nový superlektin s dvěma oddělenými cukr-vazebnými doménami a s odlišnou specifitou.

Abstract:

Lectins are carbohydrate-binding proteins of non-immune origin that bind specifically to complex carbohydrates. Some plant and animal pathogens use these protein-carbohydrate interactions in their strategy for host recognition in early stage of invasion. The presented Ph.D. thesis is approaching the study of molecular mechanisms of interactions between lectins produced by pathogenic bacteria Burkholderia cenocepacia and Ralstonia solanacearum and their carbohydrate ligands. Crystallography, surface plasmon resonance, titration microcalorimetry and other tools were used for the characterisation of two lectins. The plant bacterial pathogen Ralstonia solanacearum produces three soluble lectins RSL, RS-IIL and RS20L. All of them have been previously crystallised but the characterisation of RS20L was not complete. During this thesis, the production of the RS20L lectin was optimised and its physicochemical behaviour analysed by 2D SDS-PAGE / MALDI-MS analysis and DSC. However, different assays for determining carbohydrate specificity and affinity were not successful and different strategies have to be designed. The second part of the thesis is devoted to a lectin from the human opportunistic pathogen Burkholderia cenocepacia, responsible of high mortality in patients with cystic fibrosis or chronic granulomatous diseases. The BC2L-C lectin is a 28 kDa protein composed of two distinct domains that were separately cloned and produced in E. coli and characterised. The C-terminal domain shows sequence and structure similarity to the Pseudomonas aeruginosa lectin (PA-IIL) and recognises with high affinity D-mannosylated glycans. The N-terminal domain also displays sugar-binding ability with a strong preference for L-fucosylated oligosaccharides such as H-type and Lewis histo-blood group determinants. The N-terminal domain complexed with selenio-methyl-fucoside crystallises as a trimeric assembly that has not been observed for lectins earlier but that is highly similar to the TNF- or C1q complement structures. The BC2L-C lectin is therefore a new superlectin with two different carbohydrate-biding domains and specificities.
