Comparison of Efficiency of Tropical Wood Extraction Methods Based on Analysis of the Extract and its Influence on Wood Decaying Fungi

Abstract:

Diplomová práce se zabývá porovnáním účinnosti dvou metod extrakce: extrakční aparaturou FexIKA – extraktorem a ultrazvukem. Pro porovnání metody byly vytvořeny extrakty ze dřeva merbau (Intsia bijuga O.Ktze.) a jatoba (Hymenaea courbaril L.). Tato tropická dřeva byla rozemleta pro účely extrakce, extrahována a z extraktů byly vytvořeny tři koncentrace ochranného prostředku. Pro porovnání extrakčních metod byly ochranné prostředky testovány na obsah extraktivních látek a na obsah polyfenolických látek. Získané ochranné prostředky byly impregnovány do dřeva buku (Fagus sylvatica L.) a borovice (Pinus sylvestris L.). Impregnované bukové a borovicové tělesa byla podrobena rychlému testu ochranných prostředků proti houbám při působení outkovkou pestrou (Trametes versicolor L.) a popraškou sklepní (Coniophora puteana P. Karst.). Z dat získaných z testu byla posuzována ztráta hmotnosti těles buku a borovice. Výsledky ukázaly, že pouze tělesa impregnovaná ochranným prostředkem o koncentraci 3 vykazují dostatečnou ochranu dřeva. Analýza ochranných prostředků ukázala u dřeva merbau vyšší výtěž přístrojem FexIKA – extraktorem, u dřeva jatoba byla výtěž mírně vyšší u extrakce ultrazvukem. Obecně však tělesa impregnovaná ochrannými prostředky získanými aparaturou FexIKA – extraktorem měla nižší hmotnostní úbytky než tělesa impregnovaná ochrannými prostředky získanými ultrazvukem.

Abstract:

The thesis compares the efficiency of two extraction methods: using the FexIKA apparatus and using ultrasound. To compare the extraction methods, extracts from merbau (Intsia bijuga O.Ktze.) and jatoba (Hymenaea courbaril L.) were created. These tropical woods were ground and extracted. From the extracts, three concentrations of protective substances were created. The protective substances were tested to determine the content of extractive compounds and polyphenolic compounds. The obtained protective substances were impregnated into the wood of common beech (Fagus sylvatica L.) and pine (Pinus sylvestris L.). The impregnated beech and pine samples were tested to determine the protective effectiveness against Trametes versicolor L. and Coniophora puteana P. Karst. From the acquired data, the loss of mass of the wood was assessed. The results showed that only samples impregnated with the protective substance with concentration of 3 showed adequate protection. Analysis of the protective substances showed a higher yield when using FexIKA on Merbau wood. Jatoba wood extraction showed a slightly higher yield when extracting using ultrasound. In general, the samples impregnated with extracts obtained using FexIKA showed lower mass loss than samples impregnated with extracts obtained using ultrasound.
