Evolution and development of pharyngeal vertebrate teeth on the zebrafish model (Danio rerio)

Abstract:

Druh Danio rerio je již desítky let osvědčeným a účinným modelem pro různorodé biologické disciplíny, od evoluční biologie po fyziologii a toxikologii. Jeho významnost pramení především z detailní znalosti genetického pozadí a také ze snadné manipulace, jak z pohledu chovatelského, tak zejména z pohledu výzkumného. Důležitým znakem Danio rerio je absence orálního zubního aparátu, avšak přítomnost faryngálních zubů umístěných pouze na nejvíce posteriorním (sedmém) faryngeálním oblouku. Výzkum faryngeální dentice je velice zajímavý z hlediska evoluční biologie. Existuje totiž několik teorií popisujících evoluční vývoj dentice, mezi něž patří dvě dominantní: inside-out a outside-in, spolu s teoriemi alternativními. Hlavním důvodem pro vznik různorodých teorií je především otázka, zda zubní elementy vznikly původně z ektodermu na povrchu těla, z endodermu přítomného ve faryngu nebo z některé z jejich kombinací. Tato bakalářská práce shrnuje vlastnosti a výhody modelového organismu Danio rerio, tzv. zebřičky. Dále se věnuje shrnutím dosavadních znalostí jednotlivých teoriích evoluce vývoje faryngálních zubů. Jsou popsány některé procesy a signální dráhy hrající důležitou roli při embryonálním vývoji faryngeálních zubů. Poslední část je věnována příkladům praktického využití pro lidskou patologii, ve které má model rozsáhlá a důležitá uplatnění, jak z hlediska patogeneze, tak léčby.

Abstract:

The Danio rerio has for decades been a proven and effective experimental model for diverse biological disciplines, from evolutionary biology to physiology and toxicology. The importance of this biological models comes mainly from our detailed knowledge of its genetical background and also from the ease of manipulation, both from the breeding and scientific point of view. Danio rerio is also distinct due to being equipped with pharyngeal teeth on the posteriormost (seventh) pharyngeal arch. Research of pharyngeal dentition is important from the evolutionary viewpoint, however, the exact mechanism of it´s development is still unknown. The several theories are describing the evolution of teeth, with two of them being dominant: inside-out and outside-in, followed by other, alternative ones. The main reason for the existence of multiple theories is the question of whatever the teeth originally came from the ectoderm on the surface of the body, endoderm of pharynx or any of their combinations. This bachelor thesis summarize properties of model organism Danio rerio, also known as „zebrafish“. The further chapter is aimed at presenting multiple evolutionary theories involving pharyngeal teeth evolution. The description of evolution select processes and signal pathways involved in important parts of pharyngeal teeth development follows. The last part is focused on covering the practical aspects of using zebrafish as model organism, mainly in study of human pathology, in which the model has multiple important roles, both in pathogenesis and therapy.
