Influence of Streptococcus uberis bacteria on the proportion of CD4 positive lymphocytes of bovine mammary gland

Abstract:

Cílem této diplomové práce bylo zhodnotit vliv bakterií Stretococcus uberis na zastoupení CD4 pozitivních lymfocytů mléčné žlázy skotu. Pro experiment bylo použito deset jalovic, kříženek holštýnského a českého strakatého plemene. Jejich mléčné žlázy byly experimentálně infikovány pomocí bakterie S. uberis. Následně byly populace buněk získány výplachem mléčné žlázy po 24, 48, 72 a 168 hodinách po vyvolání infekce. Nejvyšší nárůst počtu somatických buněk byl detekován po 24 hodinách od vyvolání zánětu. Pomocí průtokového cytometru bylo zjištěno, že tento akutní zánět nejprve vyvolal aktivaci neutrofilů, jejich počet postupně klesal, a naopak se zvyšovalo zastoupení makrofágů. U subpopulace CD4 pozitivních lymfocytů bylo taktéž pozorováno postupné zvyšování procentického zastoupení v počáteční fázi zánětu. Čímž bylo prokázáno, že CD4 pozitivní lymfocyty mají významnou roli při propuknutí akutního zánětu mléčné žlázy.

Abstract:

The aim of this diploma thesis was to evaluate the effect of Stretococcus uberis bacteria on the representation of CD4 positive lymphocytes in the mammary gland of cattle. Ten heifers, Holstein crossbreeds and Czech spotted breeds were used for the experiment. Their mammary glands were experimentally infected with S. uberis. Subsequently, cell populations were obtained by irrigation of the mammary gland after 24, 48, 72 and 168 hours after infection. The highest increase in somatic cell count was detected 24 hours after inflammation. Using a flow cytometer, it was found that this acute inflammation first caused the activation of neutrophils, their number gradually decreased, and conversely, the proportion of macrophages increased. A gradual increase in the percentage in the initial phase of inflammation was also observed in the CD4 positive lymphocyte subpopulation. Thus, CD4 positive lymphocytes have been shown to play an important role in the onset of acute mammary gland inflammation.
