Application of macrocyclic compounds in separation methods

Abstract:

Tato diplomová práce se zabývá studiem cyklodextrinů, jejich inkluzních komplexů s deriváty adamantanu (kyselinou adamantankarboxylovou, amantadinem, rimantadinem a adamantylethylendiaminem) a využitím cyklodextrinů v elektro-migrační separační metodě izotachoforéze (ITP) s vodivostní detekcí. Práce je zaměřena na nenabité cyklodextriny (β-cyklodextrin, hydroxypropyl-β-cyklodextrin s průměrným stupněm substituce 4,5 a methyl-β-cyklodextrin s průměrným stupněm substituce 12) a cyklodextriny se záporným nábojem (sulfobutyl ether β-cyklodextrinu sodného s průměrným stupněm substituce 4, 6 a 10 a karboxymethyl-α/β/γ-cyklodextrin sodný s průměrným stupněm substituce 3,5). Teoretická část práce obsahuje literární rešerši o elektromigračních metodách, z nichž je podrobněji popsán princip, instrumentace a vyhodnocení izotachoforézy. V dalších kapitolách jsou shrnuty informace o struktuře, vlastnostech a širokém využití cyklodextrinů a adamantanu. Experimentální část práce je v první řadě zaměřena na ITP analýzy derivátů adamantanu v systémech bez a za přítomnosti vybraných cyklodextrinů. Dále je studováno možné využití nalezených optimálních experimentálních podmínek analýz k měření reálných vzorků: stanovení hydrogenuhličitanu v minerálních vodách a stanovení účinné látky, amantadinu, v léčivých přípravcích Viregyt-K a PK-merz používaných k léčbě Parkinsonovy choroby. Podstatná část experimentální práce je také věnována ITP analýzám aniontových derivátů β-cyklodextrinu a závěrem jsou popsány výsledky konduktometrických měření zaměřených na tvorbu inkluzních komplexů amantadinu a rimantadinu s cyklodextriny.

Abstract:

This diploma thesis deals with the study of cyclodextrins, their inclusion complexes with adamantine derivatives (adamantanecarboxylic acid, amantadine, rimantadine and adamantylethylenediamine) and the application of cyclodextrins in electromigration separation method isotachophoresis (ITP) with conductivity detection. The thesis is focused on uncharged cyclodextrins (β-cyclodextrin, hydroxypropyl-β-cyclodextrin with an average degree of substitution 4,5 and methyl-β-cyclodextrin with the average degree of substitution 12) and cyclodextrins with negative charge (sulfobutyl ether β-cyclodextrin sodium salt with the average degree of substitution 4, 6 and 10 and carboxymethyl-α/β/γ-cyclodextrin sodium salt with the average degree of substitution 3,5). The theoretical part of the thesis contains a literature review of electromigration methods, which describes in more detail the principle, instrumentation and evaluation of isotachophoresis. In the following chapters information about the structure, properties and wide use of cyclodextrins and adamantane is summarized. The experimental part of the thesis is primarily focused on ITP analyses of adamantane derivatives in systems without and with the presence of selected cyclodextrins. Furthermore, the possible utilization of the found optimal experimental conditions of the analyses for real samples measurement is studied: determination of bicarbonate in mineral waters and determination of active substance, amantadine, in Viregyt-K and PK-merz medicines used for the treatment of Parkinson's disease. A substantial part of the experimental work is also devoted to ITP analyses of anionic derivatives of β-cyclodextrin and in conclusion results of conductometric measurements focused on formation of inclusion complexes of amantadine and rimantadine with cyclodextrins are described.
