Possibilities of pharmacological regulation of mitochondrial respiration

Abstract:

Respirační rychlost mitochondrií je parametr, který charakterizuje aktivitu elektronového transportního systému (ETS) v mitochondriích a je úzce vztažen k celkové produkci ATP buňkou. Rychlost spotřeby kyslíku v izolovaných mitochondriích, uvedených do různých respiračních stavů vhodnými substráty, inhibitory a odpřahovači, lze použít pro studium změn v buněčné energetice vyvolaných xenobiotiky, jako jsou léčiva a látky obsažené v doplňcích stravy. Jako vhodný biologický model lze pro tato měření použít mitochondrie izolované ze zvířecích mozků. Cílem této práce bylo jednak nalezení optimálních experimentálních podmínek pro měření vlivu biologicky aktivních látek na respirační rychlost mitochondrií izolovaných z prasečích mozků, jednak studium vlivu vybraných látek (simvastatinu, acetylcysteinu a acetylkarnitinu) na mitochondriální respiraci. Ve vzorcích hrubé mitochondriální frakce (CMF) byl nejprve proměřen a analyzován vliv různých koncentrací substrátů a inhibitorů systému oxidační fosforylace. Poté byl změřen a analyzován vliv simvastatinu na respirační rychlost mitochondrií, přičemž byly porovnány účinky tohoto léčiva ve vzorcích CMF a v mitochondriích čištěných na sacharózovém gradientu (PMS) nebo na percollovém gradientu (PMP). Výsledky ukázaly, že všechny tři testované biologické modely (CMF, PMS, PMP) jsou dobře použitelné pro studium účinků léčiv a jiných biologicky aktivních látek na mitochondriální respiraci, ale kvantitativně přesnější výsledky poskytuje měření na čištěných mitochondriích. Vliv acetylcysteinu a acetylkarnitinu na respirační rychlost mitochondrií byl proto měřen ve vzorcích PMS. Byly zjištěny silné inhibiční účinky simvastatinu na respirační rychlost, což by mohlo souviset s některými nežádoucími účinky tohoto léku. Acetylcystein a acetylkarnitin respirační rychlost mitochondrií prakticky neovlivnily. Všechny tři testované látky (především simvastatin a acetylkarnitin) vykázaly protektivní účinky proti snížení mitochondriální respirace vyššími koncentracemi volných vápenatých iontů.

Abstract:

Respiratory rate of mitochondria is a parameter which characterizes the activity of the electron transport system (ETS) in mitochondria and is closely related to the overall production of ATP by the cell. Rate of oxygen consumption in isolated mitochondria, referred to various respiratory conditions suitable substrates, inhibitors and decoupler may be used to study changes in the cellular energy induced by xenobiotics such as drugs and substances contained in food supplements. As a suitable biological model can be used for these measurements mitochondria isolated from animal brains. The aim of this study was to find the optimal experimental conditions for measuring the impact of biologically active substances on the respiration rate of mitochondria isolated from pig brains and also study the effect of selected substances (simvastatin acetylcysteine and acetylcarnitine) on mitochondrial respiration. In samples crude mitochondrial fraction (CMF) was first measured and analyzed the effect of different concentrations of substrates and inhibitors of oxidative phosphorylation. It was then measured and analyzed the effect of simvastatin on mitochondrial respiration rate, while compared the effects of this drug in the samples and in the mitochondria CMF purified on a sucrose gradient (PMS) or percollovém gradient (PMP). The results showed that all three tested biological models (CMF PMS PMP) are well suited for studying the effects of drugs and other biologically active compounds on mitochondrial respiration, but quantitatively accurate results provides measurements of purified mitochondria. Effect acetylcysteine and acetylcarnitine mitochondria respiratory rate was therefore measured in samples of PMS. They were detected strong inhibitory effects of simvastatin on the respiration rate, which could be associated with some adverse effects of this drug. Acetylcysteine and acetylcarnitine mitochondrial respiration rate virtually had no impact. All three test compounds (particularly simvastatin and acetylcarnitine) showed protective effects against the decrease of mitochondrial respiration higher concentrations of free calcium ions.
