Detection of cancer biomarkers using immunoassays with magnetic preconcentration

Abstract:

Prostatický specifický antigen (PSA) patří mezi nejdůležitější biomarkery rakoviny prostaty. Citlivá detekce PSA je kritická pro včasnou diagnostiku, která umožňuje efektivnější léčbu. Imunochemické metody včetně enzymového imunostanovení (ELISA) patří mezi nejpoužívanější metody detekce PSA. ELISA stanovení se ale vyznačují několika nevýhodami, které je činí nedostatečně citlivými pro včasnou diagnostiku onemocnění. Foton-upkonverzní nanočástice (UCNPs) představují slibnou alternativu pro běžně používané enzymové značky v imunostanoveních. Po biokonjugaci s protilátkou nebo streptavidinem je možné UCNPs využít jako citlivou značku v imunochemických stanoveních, včetně upkonverzního imunostanovení (ULISA) využívajícího mikrotitrační destičky jako pevnou fázi. Jelikož jsou však některé biomarkery přítomné v mimořádně nízkých koncentracích, je nutné citlivost imunostanovení dále zvyšovat. Magnetické mikročástice (MBs) představují vhodnou alternativu pro mikrotitrační destičky. Mezi jejich hlavní výhody patří superparamagnetické vlastnosti, které umožňují prekoncentraci analytu, a tudíž další zlepšení citlivosti. Tato diplomová práce se zabývá využitím MBs v kombinaci s UCNPs pro vývoj imunostanovení pro detekci PSA. Biokonjugáty UCNPs s protilátkou (Ab-UCNPs) a streptavidinem (SA-UCNPs) byly charakterizovány metodami dynamického rozptylu světla (dynamic light scattering, DLS) a analýzy trajektorie nanočástic (nanoparticle tracking analysis, NTA). Jejich funkčnost byla ověřena prostřednictvím ULISA stanovení využívajících mikrotitrační destičky. Pro vývoj imunostanovení na MBs byl jako modelový analyt použitý lidský sérový albumin (HSA). Byly sledovány podmínky pro průběh imunostanovení, včetně volby optimálního pufru, koncentrace nanočástic, protokolu pro skenování dat apod. Zjištěné parametry byly použity pro další optimalizace imunostanovení pro detekci PSA. S optimálními podmínkami byla provedena analýza modelových reálných vzorků krevního séra a výsledky byly srovnány s konvenčním ELISA stanovením. ULISA stanovaní využívající MBs dosáhlo LOD 11,1 pg/ml, což byl výsledek srovnatelný s ELISA stanovením využívajícím MBs (10,2 pg/ml). Nakonec bylo provedeno stanovení s magnetickou prekoncentrací, které vedlo k citlivé detekci PSA s nejnižším dosaženým LOD o hodnotě 0,25 pg/ml, což představuje 44násobné zlepšení oproti ULISA stanovení s MBs, čímž byl demonstrován klinický potenciál vyvinuté metody.

Abstract:

Prostate-specific antigen (PSA) is one of the most important biomarkers of prostate cancer. Sensitive biomarker detection is essential for early-stage diagnosis, enabling more effective treatment. Immunochemical methods, such as enzyme-linked immunosorbent assay (ELISA), are widely used for PSA detection. However, there are several limitations that render ELISA insufficient regarding the sensitivity. Photon-upconversion nanoparticles (UCNPs) are a promising alternative to the conventionally used labels. When conjugated with biorecognition molecules, UCNPs can be used as a label in various immunoassay formats, including microtiter plate-based upconversion-linked immunosorbent assay (ULISA). However, as many biomarkers are present in extremely low levels, even ULISA may still not reach the required sensitivity, making it necessary to search for more sensitive approaches. Magnetic microparticles (MBs) represent a promising alternative for microtiter plates used in immunoassays, mainly for their superparamagnetic properties that allow for analyte preconcentration and thus higher assay sensitivity. This diploma thesis focused on the development of an immunoassay combining MBs and UCNPs for the detection of PSA. Bioconjugates of UCNPs with antibody (Ab-UCNPs) or streptavidin (SA-UCNPs) were characterized via dynamic light scattering (DLS) and nanoparticle tracking analysis (NTA) and their functionality was confirmed by microtiter plate-based immunoassays. For the development of the MB-based assays, human serum albumin (HSA) was utilized as a model analyte during the initial optimizations. Conditions for the assay procedures, including the choice of an optimal buffer, concentration of MBs, readout parameters, etc., were studied. The obtained results served for further optimizations of the immunoassay for the detection of PSA. The conditions were then applied in the analysis of model real samples of serum, which was compared to the results of the conventional ELISA utilizing both microtiter plate and MBs as solid phase. The MB-based ULISA in serum has reached the LOD of 11.1 pg/mL, which was comparable to the LOD reached in the MB-based ELISA (10.2 pg/mL). Finally, an immunoassay with an additional magnetic preconcentration step was performed, which led to sensitive detection of PSA with the lowest achieved LOD of 0.25 pg/mL, representing a 44-fold improvement compared to the MB-based ULISA, thus demonstrating the clinical potential of the developed method.
