Selection of Saccharomyces cerevisiae strain for production of beta-glucan in industry

Abstract:

Beta-glucan is a polysacharide present in the cell wall of the yeasts. Cell wall represents 30 % of dry cell mass, while glucans, which are polymers of glucosis account for 60 % of cell wall mass. Glucans are presented in cell wall of Saccharomyces cerevisiae in two forms, as beta-1,3-glucan and beta-1,6-glucan. Isolated beta-glucan has a wide range of characteristics for which is it used in the production of food supplements, medical supplies and drugs. It has also antimicrobial and antitumour effects due to its ability to modulate the immune system and to regulate blood sugar or cholesterolemia. In this diploma thesis we study a selection of S. cerevisiae strain suitable for industrial production of beta-glucan. Selection and optimization were done in the condition of submerged cultivation by determination of beta-glucan content in a dry cell mass. After selection of the strain with the highest content of beta-glucan, the cultivation conditions for growth and synthesis of beta-glucan were optimized. Batch fermentation in fermenter (bioreactor) was also performed. According to the acquired results, no significant impact of cultivation conditions and composition of cultivation medium for the beta-glucan production was observed.

Abstract:

Beta-glukán je polysacharid zastúpený v bunkovej stene kvasiniek. Bunková stena pritom predstavuje 30 % zo suchej váhy bunky a glukány ako polyméry glukózy tvoria 60 % z váhy bunkovej steny. Glukány sú v bunkovej stene kvasinky Saccharomyces cerevisiae zastúpené dvoma typmi beta-1,3-glukánom a beta-1,6-glukánom. Izolovaný beta-glukán má celú rad vlastností, pre ktoré je využívaný pri výrobe ako potravinových doplnkov, tak aj zdravotníckych prostriedkov a liečiv. Vykazuje antimikrobiálne účinky, pôsobí ako modulátor imunitného systému, ale má taktiež protirakovinové účinky a schopnosť regulovať glykémiu ci cholesterolémiu. V tejto diplomovej práci sme sa zaoberali selekciou kmeňa S. cerevisiae vhodného na využitie pre produkciu beta-gukánu v prevádzkovej mierke. Selekcia a optimalizácia prebiehala v podmienkach submerznej kultivácie stanovením obsahu beta-glukánu v sušine. Po selekcii kmeňa s najvyšším obsahom beta-glukánu boli optimalizované podmienky kultivácie pre rast a tvorbu beta-glukánu. Prevedená bola i vsádzková kultivácia vo fermentore (bioreaktore). Zo získaných výsledkov nie je zjavný výrazný vplyv podmienok kultivácie a zloženia kultivačného média na tvorbu beta-glukánu.
