The role of MSX1 gene in pathogenesis of hypodontia

Abstract:

Hypodoncie je stav vyznačující se vrozenou absencí zubů. Pokud je počet chybějících zubů do šesti nazýváme tento stav hypodoncie, chybí-li šest a více zubů jedná se již o oligodoncii. Příčinou jsou mutace ovlivňující již samotný embryologický vývoj zubu, který je řízen kaskádou dějů spouštěných několika geny, přičemž jedním z nich je i Msx1. Jsou známy tři mutace v exonu 1 a čtyři mutace v exonu 2 spojené s hypodoncií, postihující převážně druhé premoláry a třetí moláry. Gen pro Msx1 je také spojen s rozštěpem rtu a patra a Witkopovým syndromem. V této práci byly zkoumány sekvence exonu 1 a 2 a poté následně porovnány s referenční sekvencí. U sekvencí exonu 1 byly nalezeny mutace vedoucí k záměně aminokyselin, u exonu 2 se však všechny sekvence jevily jako standardní.

Abstract:

Hypodoncia is a physical state significant for inherited absence of teeth. The cases when the number of missing teeth is lower than 6 is called hypodoncia, cases of 6 and more teeth missing are already marked as oligodoncia. The cause is a mutation originating already in the embryologic development of a tooth managed by a sequence of events triggered by certain genes, one of them being the Msx1. Three mutations in exon 1 and four mutations in exon 2 related to hypodoncia affecting predominantly secondary premolar and tertiary molar teeth are currently known. The Msx1 gene is also connected with creation of a cleft lip and palate and development of Witkop syndrome. This work investigates the sequences of exon 1 and 2 and consequently compares the findings with a reference sequence. While in sequences of exon 1 mutations resulting also in change of amino acids were detected, the exon 2 sequences all seemed standard.
