Cell senescence, aging, and tumors

Anotácia:

Cílem této bakalářské práce je shrnout dosavadní poznatky o příčinách buněčné senescence, drahách její aktivace a vlastnostech senescentních buněk. Dalším cílem práce je popsat úlohu buněčné senescence ve stárnutí organizmu a ve vývoji nádorů. První část práce obsahuje historický úvod do problematiky, popis struktury a funkce telomer a mechanizmu alternativního prodlužování telomer. Další část pojednává o příčinách a mechanizmech aktivace replikativní a předčasné senescence a o charakteristice senescentních buněk. Třetí část se zabývá působením senescentní buňky na okolní tkáň a souvislostí senescence se stárnutím. Poslední část vysvětluje vztahy mezi buněčnou senescencí, stárnutím a nádory. V závěru je nastíněna možnost využití indukce buněčné senescence při léčbě nádorů.

Abstract:

The aim of this thesis is to summarize current knowledge about causes of cellular senescence, pathways of its activation and characteristics of senescent cells. The work describes the role of cellular senescence in organismal aging and cancer development. First part contains historical introduction to the subject, description of telomere structure and function and the mechanism of alternative lengthening of telomeres. Next part discusses causes of replicative and premature senescence and pathways of senescence activation. The effects of senescent cell on surrounding tissue and the relationship between senescence and aging are discussed in the third part. Last part of the text explains the link among cell senescence, aging and cancer. In conclusion, the possibility of using senescence induction in treatment of cancer is outlined.
