Abstract:

Bakterie Staphylococcus aureus je běžnou součástí mikroflóry lidí i zvířat. Jako oportunní patogen je však schopna způsobovat nejrůznější infekční onemocnění. S rostoucím výskytem bakteriálních kmenů s mnohonásobnou rezistencí k antibiotikům je třeba hledat alternativy léčby těchto infekcí. Jednou z alternativ je fágová terapie, tedy terapie pomocí bakteriofágů, které lyzují bakteriální buňky. Bakterie si však vyvinuly mechanismy rezistence i k fágům. Znalost těchto mechanismů je důležitá pro vývoj účinných fágových preparátů vhodných pro léčbu infekcí. Diplomová práce byla zaměřena na veterinární kmeny S. aureus, neboť ve veterinárním prostředí způsobují komplikace nejen bakteriální infekce zvířat, ale i kontaminace tohoto prostředí, která se pak může přenášet na lidi. Studované kmeny byly rezistentní k polyvalentním fágům, které jsou pro přípravu preparátů využívány nejčastěji. Cílem práce bylo zjistit, zda se rezistentní kmeny vyznačují společnými rysy, které by mohly za rezistenci k fágům zodpovídat. Kmeny byly charakterizovány z hlediska obsahu profágů, R-M systémů a plazmidů. Dále byla u kmenů prověřena schopnost fágů adsorbovat se na necitlivé kmeny. U kmenů byly také určeny rezistence k antibiotikům a další vlastnosti genotypu.

Abstract:

Bacteria Staphylococcus aureus is a common component of the microflora of humans and animals. As an opportunistic pathogen it is able to cause severe infectious diseases. With the growing number of multidrug resistant bacterial strains it is important to find alternatives for the treatment. One of these is phage therapy, meaning therapy with bacteriophages which lyse the bacterial cells. Unfortunately, bacteria also evolved mechanisms of resistance to phages. The knowledge of these mechanisms is important for the development of effective phage preparations which are used to treat infections. This diploma thesis was focused on veterinary strains of S. aureus because in the veterinary environment they cause troubles not only with animal infections, but they also contaminate the surroundings and can spread to humans. The studied strains were resistant to polyvalent phages which are most frequently used for the phage preparations. The goal of the thesis was to find out whether these phage-resistant strains carry common features which could cause the resistance. The strains were characterized in terms of the content of prophages, R-M systems and plasmids. Then the ability of phages to adsorb to cells was studied; also resistance to antibiotics and other genotype properties were determined.
