Novel techniques in evaluation of endocrine disrupting compounds in contaminated waters

Abstract:

Řada sloučenin, které vstupují do vodního prostředí v důsledku antropogenních aktivit, může způsobit narušení endokrinního systému živočichů i člověka. Jelikož tyto sloučeniny mohou napodobovat přirozené hormony, měnit jejich fyziologické koncentrace nebo narušovat syntézu/metabolismus hormonů v exponovaných organismech, tak je jejich sledování a omezování v matricích životního prostředí velmi důležité pro zachování integrity biologické rozmanitosti. Tyto tzv. endokrinní disruptory (EDCs – endocrine disrupting compounds) mohou vstupovat do přírodních vod z různých bodových a difúzních zdrojů, jako jsou čistírny odpadních vod, průmyslové zdroje, odtoky zemědělských ploch atd. a mohou zahrnovat velké množství sloučenin od pesticidů po výrobky osobní péče. Studie účinků látek narušujících endokrinní systém včetně feminizace druhů ryb a kolapsu populace ryb, jakož i detekce velkého množství chemických látek ve vodním prostředí, vedly k požadavkům na monitoring EDCs ze strany státu či agentur na ochranu životního prostředí. V reakci na to jsou intenzivně vyvíjeny metody pro hodnocení typů, koncentrací či účinků EDCs. Citlivost a robustnost metod instrumentální analýzy a biologických účinků hodnocení se zlepšuje a nyní jsou schopny detekci koncentrace velkého množství sloučenin při nízké koncentraci ng/l. Vzhledem k jistým výhodám jsou dnes in-vitro biotesty stále více přijímány pro hodnocení účinků EDCs jako vhodná alternativa chemických analýz. In-vitro biotesty se používají k měření různých způsobů endokrinní disrupce, např. estrogenní, androgenní a progestagenní aktivity v různých matricích prostředí. Účinnost in-vitro analýzy je však ovlivněna metodami přípravy a zpracování vzorků. Jelikož účinná koncentrace sloučenin je často pod detekčním limitem in-vitro biotestů, tak jsou vzorky běžně zakoncentrovávané extrakcí na pevné fázi (SPE – solid phase extraction). To však vede k separaci EDCs ze složité směsi vzorků prostředí, čímž se však také eliminuje matricový účinek, který může být buď aditivní nebo inhibiční. Proto se předpokládá, že metody přípravy vzorků, které nevedou k izolaci sloučenin od matrice, mohou poskytnout užitečnější informace o skutečných účincích na životní prostředí. Jednou z takových metod může být reverzní osmóza (RO – reverse osmosis), která se nyní běžně používá pro koncentraci produktů v potravinářském průmyslu. Cílem dizertační práce bylo vyhodnotit použitelnost RO pro zakoncentrování vzorků vody pro následnou in-vitro analýzu. K zhodnocení vhodnosti RO ke koncentraci estrogenních látek byla provedena optimalizace procesu umožňující rychlé a efektivní zpracování vzorků. Matematický model popisující tento proces byl vyvinuty v programu MATLAB-Simulink a následně byl ověřen reálnými výsledky z chemické analýzy. Dále byla studována vhodnost použití RO při in-vitro analýze estrogenních látek ve vzorcích vody z životního prostředí. In-vitro biotest s buňkami hERα-HeLa-9903 byl optimalizován pro detekci environmentálně relevantních koncentrací v zakoncentrovaných vodných vzorcích. Modelové sloučeniny zahrnovaly 17β-estradiol, 17α-etinylestradiol, estron a estriol. Teoretický koncentrační faktor RO se pohyboval v rozmezí 6 až 10 a příslušná výtěžnost RO byla považována jako přijatelná pro následnou in-vitro analýzu. Získané výsledky naznačují, že RO představuje slibnou metodou zpracování vzorků pro následnou in-vitro analýzu, která však má jak výhody, tak i nevýhody ve srovnání se standardně používanou SPE.

Abstract:

Many compounds that enter the aquatic environment due to anthropogenic activities can cause endocrine disruption of the wildlife and humans. Since endocrine disrupting compounds can mimic natural hormones and/or unbalance their physiological concentrations or interfere with hormone synthesis/metabolism in exposed organisms, their management in the environmental matrices are of great importance to preserve the integrity of ecological biodiversity. These EDCs may enter the natural waters from various point and diffused sources such as wastewater treatment plant, industrial discharge, agricultural surface runoffs etc. and may include a vast number of compounds from pesticides to personal care products. The studies on effect of endocrine disrupting compounds including feminization of fish species and collapse of a fish population as well as detection of multitude of chemicals in the aquatic environment prompted stringent monitoring and assessment requirements by the state and environmental protection agencies. In response to this, the methods used to evaluate the type and concentration as well as effects of endocrine disrupting compounds in the aquatic environment has gained momentum over the past few years. The sensitivity and robustness of instrumental analysis and biological assessment methods have improved to detect the concentration of multitude of compounds at a low concentration of ng/L. Owing to its relative advantages, in-vitro bioassays are widely accepted and used for assessing the effects of endocrine disrupting compounds in the environmental matrices. In-vitro bioassays are used in measuring various endocrine-disrupting modes of action, e.g. estrogenic, androgenic and progestogenic activities in various environmental matrices. The efficacy of subsequent in-vitro analysis is affected by sample preparation and processing methods. Since the effective concentration of the compounds are below the detection limit of the bioassays, the samples are invariably enriched commonly by solid phase extraction. This results in the isolation of the endocrine disrupting compounds from the complex mixture of the environmental samples, thus eliminating the matrix effect that might either be additive or inhibitive. Therefore, it was hypothesized that sample preparation methods which will not result in isolating the compounds might provide more useful information on the actual effects in the environment. One such method would be the reverse osmosis which is commonly adopted for concentrating products in the food industries. Therefore, the aim of the study was to evaluate the usefulness of reverse osmosis process to enrich the water samples for subsequent in-vitro analysis using bioanalytical methods. To evaluate the usefulness of reverse osmosis to concentrate estrogenic compounds, process optimization to enable rapid and effective processing of the samples was carried out. Mathematical models were developed and run in MATLAB-Simulink and the model was verified by data from the chemical analysis. The applicability of the reverse osmosis process and its usefulness in measuring estrogenic compounds in the environmental samples using in-vitro assays were studied. The in-vitro assay using hERα-HeLa9903 was adapted to conform with the expected concentration of the model compounds in the samples. The model compounds included 17-beta estradiol, 17-alpha ethinyl estradiol, estrone and estriol. The theoretical concentration factor from the reverse osmosis enrichment process ranging between 6 to 10 was deemed acceptable for in-vitro measurement. Overall, the findings from the thesis indicates that the reverse osmosis is a useful method of sample processing for subsequent in-vitro analysis offering relative advantages and disadvantages compared to solid phase extraction.
