Phototoxicity

Abstract:

Fototoxicita je běžná kožní reakce vznikající po aplikaci fotosenzitivních látek a následné expozici pokožky slunečnímu záření. Na ochranu před škodlivými účinky slunečního světla způsobených ultrafialovým zářením se v běžné praxi využívají produkty obsahující UV filtry. V dnešní době se však v souvislosti s využíváním běžně dostupných UV filtrů často mluví s negativními dopady, jak na organismus, tak na životní prostředí. Teoretická část diplomové práce pojednává právě o fototoxicitě, jejím mechanismu a klinických projevech. Dále je popsáno testování fototoxicity, fototoxické látky využívané v kosmetickém průmyslu a využití fototoxického účinku v medicíně. Další část teorie se zabývá oxidativním stresem, který může být způsoben právě fototoxickým účinkem na organismus. Konkrétně se zaměřuje na reaktivní formy kyslíku a důsledky oxidativního stresu. V rámci praktické části byly připravené koloidní suspenze vykazující antioxidační vlastnosti, a to polyanilinová a polypyrrolová koloidní suspenze za využití polyvinylpyrrolidonu jako stabilizátoru. Následovalo určení jejich vlastností, jako je průměrná velikost částic, index polydisperzity a cytotoxicita. Nakonec proběhlo testování fototoxicity vybraných fototoxických esenciálních olejů a vlivu připravených koloidů na jejich fototoxický účinek.

Abstract:

Phototoxicity is a common skin reaction that occurs after the application of photosensitizers and subsequent exposure of the skin to sunlight. Products containing UV filters are commonly used to protect against the harmful effects of sunlight caused by ultraviolet radiation. Today, however, in connection with the use of commonly available UV filters, there are often discussions about the negative effect on the organism and the environment. The theoretical part of the diploma thesis is about phototoxicity, it's mechanism and clinical manifestations. Next, phototoxicity testing, phototoxic substances in the cosmetic industry and the use of the phototoxic effect in medicine are also described. The next part of the theory is about oxidative stress, which can be caused by a phototoxic effect on the organism. Specifically, it focuses on reactive oxygen species and the consequences of oxidative stress. In the practical part, a colloidal suspension showing antioxidant properties was prepared, namely polyaniline and polypyrrole were prepared in the presence of polyvinylpyrrolidone as a stabilizer. This was followed by the determination of their characteristics such as average particle size, polydispersity index and cytotoxicity. Finally, the phototoxicity of selected essential oils and the effect of prepared colloids on their phototoxic impact were tested.
