Abstract:

Tato diplomová práce se zabývá charakterizací karyotypu tribu Cardamineae (Brassicaceae) s využitím cytogenetických metod založených na fluorescenční in situ hybridizaci. V první části byla pomocí metody komparativního chromozomální paintingu objasněna karyotypová struktura vybraného druhu Cardamine hirsuta (n=8) z rodu Cardamine. Ve druhé části byl s využitím genomické in situ hybridizace objasněn alopolyploidní původ tetraploidního druhu Cardamine flexuosa (2n=4x=32) z rodu Cardamine a autopolyploidní původ oktoploidního druhu Nasturtium microphyllum (2n=8x=64) z rodu Nasturtium. Struktura karyotypu druhu C. hirsuta je velmi podobná ancestrálnímu karyotypu čeledi Brassicaceae (n=8). Během evoluce v ní došlo k reciproké translokaci mezi krátkými rameny ancestrálních chromozomů AK6 a AK8 zahrnující konzervované chromozomové oblasti O a P (AK6) a V (AK8). Tato struktura karyotypu potvrzuje velmi blízkou příbuznost rodu Cardamine (tribu Cardamineae) s tribem Camelineae, do kterého jsou zařazeny druhy s ancestrální sestavou chromozomů. Pro objasnění předpokládané ancestrální struktury karyotypu zástupců tribu Cardamineae ovšem bude nezbytné provést další KCP u jiných druhů z tohoto tribu. Při studiu polyploidních druhů z tribu Cardamineae byl prokázán alopolyploidní původ druhu C. flexuosa, u kterého byly rozpoznány dva rodičovské genomy – C. amara (2n=16) a C. hirsuta (2n=16). U polyploidního druhu N. microphyllum byl objasněn jeho vznik autopolyploidizací genomu N. officinale (2n=32).

Abstract:

This diploma thesis deals with characterization of the karyotype structure of the tribe Cardamineae (Brassicaceae) using cytogenetic methods based on the fluorescence in situ hybridization. In the first part, karyotype structure of Cardamine hirsuta (n=8) from the genus Cardamine was clarified by comparative chromosome painting (CCP). In the second part, polyploid origins of tetraploid Cardamine flexuosa (2n=4x=32) from the genus Cardamine and octoploid Nasturtium microphyllum (2n=8x=64) from the genus Nasturtium were illuminated by genomic in situ hybridization. Karyotype structure of C. hirsuta was resembling to the ancestral karyotype of the familly Brassicaceae (n=8). During the evolution, one reciprocal translocation between two short arms of the chromosomes AK6 and AK8 including conservative building blocks O and P (AK6) and V (AK8) occurs. This karyotype structure confirms close relationship of the genus Cardamine (tribe Cardamineae) and the tribe Camelineae in which species with ancestral karyotype include, and thereby it includes the tribe Cardamineae into the clade heading into the ancestral karyotype. For clarifying supposed ancestral structure of species from the tribe Cardamineae, it is necessary to use CCP on another species from this tribe to evaluate their karyotype structure. During the study of polyploid species of the tribe Cardamineae, allopolyploid origin of C. flexuosa was demonstrated; two parental genomes were recognized – C. amara (2n=16) and C. hirsuta (2n=16). Autopolyploid origin by diploidization of the genome of N. officinale (2n=32) was illustrated in N. microphyllum.
