Modern analysis of molecular data using R and Bioconductor.

Abstract:

Studium exprese genů, jakožto jedno z hlavních dogmat molekulární biologie, hraje klíčovou roli při pochopení funkčnosti biologických procesů. S příchodem vysokopokryvných metod se vědcům otevřely dveře k rozsáhlému zkoumání této problematiky. Mezi takové metody můžeme zařadit například mikročipy, kvantitativní polymerázovou řetězovou reakci a dvourozměrnou gelovou elektroforézu. V této práci jsem sepsal stručný úvod do těchto metod, provedl základní analýzu zabývající se hledáním rozdílů v genové expresi a rovněž popsal problematiku tohoto testování. Data byla zpracována a vyhodnocena Bioconductorem využívající prostředí R. Náplní práce je předvést užitečnost tohoto programu na poli molekulární biologie.

Abstract:

Study of gene expression, as one of the main dogma of molecular biology, plays a crucial role in understanding the functionality of biological processes. With arrive of high-throughput methods scientists have opened the door to extensive exploration of this issue. These methods can include for example microarrays, quantitative PCR and two-dimensional gel electrophoresis. In this paper, I wrote a brief introduction to these methods, performed basic analysis dealing with the search for differences in gene expression and also described the problems of this testing. Data were processed and evaluated using Bioconductor with R environment. The aim of this work is to demonstrate utility of this program on the field of molecular biology.
