Hydrogels for in vitro tissue models

Abstract:

Cílem diplomové práce je připravit hydrogely pro in vitro model tenkého střeva. Teoretická část práce se zabývá popisem vybraných biomakromolekul mezibuněčné hmoty (kolagenu, fibronektinu, kyseliny hyaluronové) a popisem různých typů tkáňových modelů, především těch specifických pro tenké střevo. Experimentální část práce zahrnuje vytvoření kompozitního hydrogelu kombinujícího kolagen, thiolovanou kyselinu hyaluronovou a fibronektin a následnou charakterizaci hydrogelu. Charakterizace materiálu byla provedena jednak měřením viskoelastických vlastností pomocí reometru a dále stanovením vybraných parametrů buněčné kompatibility s využitím buněčné linii myších embryonálních fibroblastů NIH/3T3. Stanovena byla míra cytotoxicity pomocí dvou postupů, a to cytotoxicity extraktu a cytotoxicity v přímém kontaktu. Následně byla stanovena schopnost buněk růst a množit se uvnitř hydrogelu, a to pomocí konfokální mikroskopie.

Abstract:

The aim of this thesis is to prepare hydrogels for an in vitro model of the small intestine. The theoretical part of the thesis focuses on the description of collagen, fibronectin, hyaluronic acid and different types of tissue models, especially those for the small intestine. The experimental part of the work involves the creation of a composite hydrogel combining collagen, thiolated hyaluronic acid and fibronectin. The characterization of the material was performed by measuring the viscoelastic properties using a rheometer. The biological properties of the hydrogel were also tested on the NIH/3T3 mouse embryonic fibroblast cell line, including cytotoxicity testing of the extract and in direct contact. Cell proliferation inside the hydrogel was monitored by confocal microscopy.
