Health status of amphibians in Drahany Highlands and its surroundings

Abstract:

Batrachochytrium salamandrivorans je smrtelný patogen ohrožující ocasaté obojživelníky způsobující mločí chytridiomykózu, který se z Asie během posledních 10 let rozšířil i do evropských zemí. Cílem této práce bylo zjistit, zda se s ním můžeme setkat i na území České republiky, konkrétně v oblasti Drahanské vrchoviny a jejího okolí. Kromě Bsal je práce zaměřena i na jiné patogeny obojživelníků, jako příbuzný Batrachochytrium dendrobatidis způsobující žabí chytridiomykózu, ranavirus a herpesvirus. K analýze byla použita environmentální DNA získaná ze vzorků vody z vybraných toků, DNA získaná prostřednictvím kožních stěrů z obojživelníků a jako metoda byla využita kvantitativní polymerázová řetězová reakce (qPCR) a LAMP. Toky byly zvoleny podle mapy výskytu mloka skvrnitého a dalších obojživelníků na nálezové databázi ochrany přírody Agentury ochrany přírody a krajiny České republiky. Testováním bylo zjištěno, že se ve vybraných potocích na území Drahanské vrchoviny a jejího okolí žádný ze zkoumaných patogenů nevyskytuje. Součástí práce je také statistické zhodnocení rozdílu ve výskytu testovaných druhů obojživelníků mezi lokalitami v oblasti Drahanské vrchoviny v jednotlivých ročních obdobích.

Abstract:

Batrachochytrium salamandrivorans is a deadly pathogen threatening the Caudata group of amphibians, causing chytridiomycosis, which has expanded from Asia to European countries over the past decade. The aim of this study was to determine whether this pathogen is also present in the Czech republic, specifically in the Drahany Highlands region and its surroundings. In addition to Bsal, the study also focuses on other amphibian pathogens, such as the related Batrachochytrium dendrobatidis causing amphibian chytridiomycosis, ranavirus and herpesvirus. Environmental DNA obtained from water samples from selected streams, DNA obtained through skin swabs from amphibians, and the method quantitative polymerase chain reaction (qPCR) and LAMP were used for the analysis. The streams were chosen based on map of the occurence of the fire salamander and other amphibians in the database of Nature Conservation Agency of the Czech republic. Testing revealed that none of the examined pathogens were present in the selected streams in the Drahany Highlands region and it's surroundings. Part of the thesis also includes a statistical evaluation of the differences in the occurrence of the tested amphibian species between locations in the Drahanská Highlands during different seasons.
