Influence of curcuminoids and its analogs to amount and function of protein p53 in tumor cell lines

Abstract:

Diplomová práce pojednává o vlivu kurkuminoidů a jejich syntetických analogů na lidské nádorové buňky. Patří mezi ně kurkumin, demethoxykurkumin, bisdemethoxykurkumin a jejich syntetické analogy např. EF-24 nebo FLLL31. Práce popisuje jejich pozitivní terapeutické vlastnosti, zejména vlastnosti proti-nádorové. Podrobně je zde rozebrán jejich vliv na molekulární úrovni, který je také v práci experimentálně zkoumán na nádorových liniích rakoviny tlustého střeva HCT 116 a rakoviny prsu MCF7. Je studován jejich vliv na míru apoptózy v nádorových buňkách a srovnána jejich proti-nádorová účinnost. Dále je zkoumána jejich schopnost působit na nádorový supresorový protein p53, a to zejména na úrovni jeho exprese a vazby k DNA-cílové sekvenci. Nakonec se lze dozvědet něco o jejich vlivu na buněčnou lokalizaci proteinu p53 v nádorových buňkách.

Abstract:

The diploma thesis discuss the effect of curcuminoids and their synthetic analogues on human cancer cell lines. They consist of curcumin, demethoxycurcumin, bisdemethoxycurcumin and their synthetic analogues, i.e. EF-24 or FLLL31. The thesis describes their positive therapeutic properties, especially their anti-tumor properties. Their effects are analyzed in detail on molecular level, which have been also examined experimentally on colorectal carcinoma HCT 116 cell line and breast cancer MCF7 cell line. We studied their effect on amount of apoptotic cancer cells and compared their anti-tumorous effectiveness. The aim of the thesis is to discover, if these therapeutics affect the tumor suppressor protein p53, especially at its expression level. Furthermore, was examined influence of these therapeutics to binding of p53 protein on its DNA-target sequence. And finally, the effect of therapeutics on the cell localization of p53 protein in cancer cells have been studied.
