Study of host-parasitoid interactions in a temperate region

Abstract:

Parazitoidi jsou nesmírně diverzifikovanou a ekologicky významnou skupinou hmyzu, přičemž trofické sítě mezi rostlinami, hmyzími herbivory a parazitoidy zahrnují většinu suchozemské biodiverzity. Studium tritrofních interakcí je tedy logickým krokem k pochopení vzniku globální diverzity hmyzu, jakým způsobem je udržována a jaké faktory ovlivňují strukturu trofických sítí. Předložená práce se opírá o celkem tři autorské publikace, z nichž dvě se zabývají vybranými aspekty ekologie a evoluce parazitoidů a jejich herbivorních hostitelů v lesích temperátní oblasti, a jedna je metodologická, řešící přístupy k identifikaci trofických interakcí mezi parazitoidy a jejich hostiteli. První práce se zabývá vlivem fylogeneze hostitelských rostlin na strukturu trofických sítí mezi rostlinami a herbivorními gildami s různou mírou specializace. Naše výsledky naznačují, že hostitelská fylogeneze má při utváření struktury trofických sítí mezi rostlinami a herbivorním hmyzem zásadní význam, přičemž záleží na stáří divergence jednotlivých hostitelských taxonů. V případě generalistů ("leaf chewers") hrají roli v utváření struktury trofických sítí hlubší linie hostitelské fylogeneze, kdežto vysoká míra specializace téměř výhradně monofágních gild ("leaf gallers", "leaf miners") je ovlivněna především terminálními uzly rostlinné fylogeneze a neodráží tedy její hlubší linie. Druhá práce se zaměřuje na stratifikaci hmyzích herbivorů ("leaf chewers") a jejich parazitoidů podél vertikálního gradientu korunového patra temperátního lesa. Navzdory obecnému paradigmatu o slabé vertikální stratifikaci členovců v lesích mírného pásma jsme zjistili signifikantní trendy v abundanci a diverzitě hostitelů a lišila se také míra parazitace, pravděpodobně vlivem proměňujících se (a)biotických podmínek. Zjistili jsme také výrazné vertikální trendy v abundanci u jednotlivých hostitelských gild ("exposed" vs. "semi-concealed feeders") a také rozdíly ve vertikálních preferencích jednotlivých taxonů parazitoidů s různou mírou specializace. Vertikální gradient se však nepromítl do struktury trofických sítí, kde jsou pravděpodobně důležitější ekologické charakteristiky jednotlivých druhů stromů. Třetí práce primárně testuje potenciál nového protokolu pro DNA metabarcoding při identifikaci interakcí mezi parazitoidy a jejich hostiteli. Posuzovali jsme také úplnost a použitelnost lokální referenční knihovny pro identifikaci jednotlivých studovaných taxonů. Studie zároveň podrobně diskutuje výhody a nevýhody jednotlivých identifikačních metod (DNA metabarcoding, barcoding a morfologická identifikace). Použitý protokol pro DNA metabarcoding se ukázal jako velmi vhodný pro přesnou a rychlou identifikaci trofických interakcí mezi herbivory a jejich parazitoidy a společně s rostoucími referenčními knihovnami může být v budoucnu užitečnou identifikační metodou v rozsáhlých ekologických studiích.

Abstract:

Parasitoids represent an extremely diverse and ecologically important group. Plant-herbivore-parasitoid food webs represent a major component of global terrestrial biodiversity. Therefore, study of tritrophic interactions is a logical step towards understanding the origin of global insect diversity, how it is maintained, and what factors shape the structure of trophic networks. This thesis is based on three publications with my co-authorship. Two of them are focused on selected aspects of ecology and evolution of parasitoids and their herbivore hosts in the temperate forests, and one is methodological, focusing on the approaches to the identification of host-parasitoid interactions. The first publication deals with the influence of host plant phylogeny on the structure of trophic networks between plants and herbivorous guilds with different degree of specialization. Our results demonstrate the high importance of host plant phylogeny in modulating plant-herbivore food web structure, with the relative importance depending on the age of divergence of individual host taxa. In the case of generalist guilds (leaf-chewers), it is the phylogeny of deeper plant lineages that drives the food web structure whereas the specialization of monophagous guilds (gallers, miners) is affected mainly by the terminal parts of the plant phylogeny and do not generally re?ect deeper host phylogeny. The second publication is focused on the vertical stratification of host-parasitoid interactions in a temperate forest canopy. Despite the general paradigm of weak arthropod stratification in temperate forests, we found significant stratification of host abundance, diversity and parasitism rates, probably resulting from changing (a)biotic conditions along a vertical canopy gradient. Moreover, we found significant vertical patterns in abundance between exposed and semi-concealed hosts, and differences in vertical preferences of individual parasitoid taxa in relation to their phylospecificity. However, the vertical gradient seems to be less of a determinant for food web structure. In such case, the ecological characteristics of individual tree species are probably more important. In the third paper, we primarily test for the potential of the new DNA metabarcoding protocol for identifying host-parasitoid interactions. Moreover, we assessed the completeness and usability of a local reference library for identification of individual study taxa. We also extensively discuss pros and cons of individual identification methods (DNA metabarcoding, standard barcoding, morphology). The protocol used for DNA metabarcoding has showed a potential to accurately determine trophic interactions between herbivores and their parasitoids, and along with growing reference libraries can be an ideal method for sample identification in the future large-scale ecological studies.
