Reliability of stable isotopes for detecting terrestrial gastropod diet

Abstract:

Suchozemští plži jsou často velmi početnou skupinou mnoha ekosystémů, kde hrají zásadní roli v potravních řetězcích. Ačkoliv se jedná o vhodnou modelovou skupinu živočichů, potravní nároky jsou stále prozkoumány jen u několika málo čeledí. Analýza stabilních izotopů představuje vhodný nástroj pro zjišťování trofických úrovní a potravních preferencí měkkýšů. Cílem praktické části této diplomové práce bylo zjistit, zda se se izotopový signál v tkáni plžů mění podle způsobu fixace analyzované tkáně plže. Bylo zjištěno, že nejlepší metodou, jak zpracovat a uchovat vzorky je usušení či zmrazení tkání. Etanol měl vliv na izotopový signál δ13C, přičemž byl vzorek vlivem této látky obohacen o 13C oproti usušení a zmrazení. Dále byly analýzou stabilních izotopů změřeny hodnoty přirozeně se vyskytujících izotopů (δ13C a δ15N) ve dvaceti vzorcích tkání plže Lissachatina fulica. Plži rozděleni do skupin po pěti jedincích byli během experimentu krmeni odlišným typem potravy, a to salátem, suchým javorem, blešivcem a směsí předchozích potrav s čerstvým žampionem. Dle výsledků byla variabilita v rámci skupin menší než variabilita mezi skupinami, nejvyšší hodno-ty δ15N byly naměřeny u skupiny na blešivci, následovala skupina krmená směsí a žampionem, ačkoliv na základě analýzy nebyl plži konzumován. Naopak nejnižší hodnoty vykazovaly jedinci skupiny krmené suchým javorovým listem. Lissachatina fulica je potravní generalista. Stravu tohoto plže tvoří převážné cévnaté rostliny, ale pokud má tento druh k dispozici živočišnou potravu, dává ji na základě výsledků zřetelně přednost. Experiment jasně prokázal, že plži odráží izotopový signál konzumované potravy. Při porovnání hodnot s hodnotami z experimentu z bakalářské práce s druhem Subulina octona je patrné, že S. octona, jakožto menší z druhů, má tkáň více obohacenou o těžší izotopy. Velikost trofických elips byla mezi všemi skupinami srovnatelná, pouze u skupin S. octona krmených blešivcem a směsí potrav s žampionem byly elipsy větší a od ostatních se lišily. Porovnáme-li skupiny obou druhů krmené všemi typy potrav a žampionem, bylo zjištěno, že menší z druhů (S. octona) má větší trofickou elipsu oproti L. fulica. Tyto výsledky jsou odpovídají datům z terénní studie, kdy větší druhy měly menší trofickou niku, jelikož oportunistické krmení vede k podobným izotopovým signálům mezi jedinci. Naopak menší druhy zřejmě obsazují kvůli omezené pohyblivosti různé mikrohabitaty lišící se potravní nabídkou, což vede k odlišným izotopovým signálům mezi jedinci. V této studii však u menších druhů byla velikost elipsy způsobena spíše odlišnými potravními preferencemi jedinců.

Abstract:

Land snails are often a very numerous organisms in many ecosystems, where they play a vital role in food chains. Although this is a suitable model group of animals, their feeding requirements have been investigated only for a few families. Stable isotope analysis is a suitable tool for determining the trophic levels and food preferences of molluscs. The aim of the practical part of this master thesis was to find out whether the isotopic signal in snail tissue changes according to the method of fixation of the analyzed snail tissue. It has been found that the best method to process and preserve the samples is to dry or freeze the tissues. Ethanol had an effect on the δ13C isotopic signal, while the sample was enriched by 13C due to this substance compared to drying and freezing. Furthermore, the values of naturally occurring isotopes (δ13C and δ15N) in twenty tissue samples of the snail Lissachatina fulica were measured by stable isotope analysis. The snails, divided into groups of five individuals, were fed a different type of food during the experiment, namely lettuce, dry maple leaves, dry Gammarus sp. and a mixture of all these food items along with fresh mushrooms. According to the results, the variability within the groups was smaller than the variability between the groups, the highest values of δ15N were measured in the group fed with Gammarus sp., followed by the group fed with the mixture and mushroom, although, based on the analysis, snails did not consumed the latter. On the contrary, the individuals of the group fed with dry maple leaf showed the lowest values. Lissachatina fulica is a food generalist. The diet of this snail consists mainly of vascular plants, but when this species has animal food available, it clearly preferred it based on the results. The experiment clearly demonstrated that snails reflect the isotopic signal of the food consumed. When comparing the values with the values from the experiment from the bachelor's thesis with the species Subulina octona, it is evident that this species, as the smaller of these species, has tissue more enriched with heavier isotopes. The size of the trophic ellipses was comparable between all groups, only in the groups of S. octona fed with Gammarus sp. and a mixture of food items with mushroom, the ellipses were larger and different from the others. If we compare groups of both species fed with all types of food and mushrooms, it was found that the smaller of the species (S. octona) has a larger trophic ellipse compared to L. fulica. These results resemble the data of a field study where larger species have a smaller trophic niche, as opportunistic feeding leads to similar isotopic signals between individuals. In contrast, a smaller species occupy different microhabitats with different food choices and their limited mobility leads to different isotopic signals between individuals. However, in this study, in the smaller species, the size of the ellipse was rather due to different food preferences of the individuals.
