Analysis of feline sera for the detection of chosen zoonotic diseases

Abstract:

Kočka domácí (Felis catus) patří bezesporu k nejoblíbenějším domácím mazlíčkům. Kočka může být napadena různými původci onemocnění. Počet koček roste a s ním i možné riziko přenosu nákazy z kočky na člověka. V předkládané diplomové práci je v teoretické části rešeršně zpracována problematika vybraných infekčních agens (epidemiologie, etiologie, patogeneze a klinické projevy) v kontextu výskytu u koček a lidí v České republice, ale i ve světě. Byly zpracovány následující nákazy: toxoplazmóza (původce Toxoplasma gondii), Lymeská borrelióza (původce Borrelia burgdorferi sensu lato), Q horečka (původce Coxiella burnetii) a tularémie (původce Francisella tularensis). Nejen tato vybraná patogenní agens způsobují zoonotická onemocnění, neboli zoonózy, která jsou přenosná ze zvířat na člověka. V experimentální části práce byla zkoumána prevalence u souboru 357 koček na všechny čtyři vybrané patogenní mikroorganismy. Všechna krevní séra pocházela od pacientů z Kliniky chorob psů a koček, Veterinární a farmaceutické univerzity v Brně. Pro detekci přítomnosti specifických protilátek IgM a IgG v kočičích sérech byla využita metoda ELISA. Největší séropozitivita koček se ukázala pro patogeny Toxoplasma gondii a Borrelia burgdorferi sensu lato, následována Coxiellou burnetii a nejméně koček bylo pozitivních na protilátky proti Francisella tularensis. Dále práce odhalila poměrně velké procento koinfekce – nákazy 2 a více patogeny současně.

Abstract:

Domestic cat (Felis catus) is one of the most popular pet. Cats can be attacked by various pathogenic microorganisms. The number of cats increase and, with it, the risk of transmission of cat-to-human infection. The theoretical part of this thesis deals with the issue of selected infectious agents (epidemiology, etiology, pathogenesis and clinical manifestations) in the context of cats and humans in the Czech Republic as well as in the world. The following infections have been described: toxoplasmosis (Toxoplasma gondii), Lyme borreliosis (Borrelia burgdorferi sensu lato), Q fever (Coxiella burnetii) and tularaemia (Francisella tularensis). But not only these selected pathogenic agents cause zoonotic diseases, which are transmissible from animal to human. The experimental part of the study examined the prevalence of 357 cats on all four selected pathogenic microorganisms. All blood sera came from patients from the Clinic of Dog and Cat Diseases, the Veterinary and Pharmaceutical University in Brno. The ELISA method was used to detect the presence of specific IgM and IgG antibodies in feline sera. The largest number of cats were positive for pathogens Toxoplasma gondii and Borrelia burgdorferi sensu lato, followed by Coxiella burnetii and Francisella tularensis. Study also revealed a relatively high percentage of coinfection - infection by 2 or more pathogens at the same time.
