Evolutionary mechanisms affecting the fixation of specific rates of aging in sexually reproducing organisms

Abstract:

Stárnutí, jenž ovlivňuje téměř každý aspekt zdraví a nemoci, je jedním z nejdůležitějších biologických procesů vůbec. Navzdory jeho důležitosti je však naše porozumění toho, jak se stárnutí vyvinulo, značně limitované. Vědci zkoumající evoluci stárnutí jsou rozdělení do dvou zdánlivě nesmiřitelných táborů a současné teorie vysvětlující evoluci stárnutí nejsou schopné odpovědět ani na základní otázku proč se stárnutí vyvinulo u většiny, ale ne u všech organizmů. Tahle práce předkládá kritický přehled současných evolučních teorií stárnutí obou znepřátelených názorových směrů a nabízí alternativní pohled, který netrpí nedostatky současných teorií. Praktická část obsahuje výsledky třech různých multiagentních modelů a výsledky analýzy lidských mortalitních dat za účelem porovnání rychlosti stárnutí mužů a žen. Celkem tahle práce předkládá pět hlavních výsledků. Prvním je, že za předpokladu, že nestárnutí vyžaduje koordinovanou interakci několika dobře vyladěných molekulárních drah a stárnutí nikoliv, stárnoucí jedinci lehce převládnou nad nestárnoucími v sexuálně se rozmnožující populaci s asortativním výběrem. Druhým zjištěním je, že nestárnutí je mnohem výhodnější pro simultánní hermafrodity než pro organizmy s odlišenými samci a samicemi. Třetí hlavní zjištění je, že v případě existence funkčního vztahu („trade-offu“) mezi důležitým znakem jako je například reprodukce a rychlostí stárnutí, je tenhle funkční vztah hlavním faktorem určujícím směřování evoluce stárnutí. Čtvrté zjištění je, že pravděpodobně existuje optimální délka života, která vede k oslabení genetického driftu. Pátým hlavním zjištěním je, že i když mají muži od narození vyšší pravděpodobnost smrti než ženy, rychlost stárnutí mužů a žen je přibližně stejná. Dohromady tahle zjištění odpovídají na mnohé otázky a nabízí tak nové příležitosti pro výzkum mechanismů ovlivňujících evoluci specifických rychlostí stárnutí.

Abstract:

Aging is arguably one of the most important biological processes affecting almost every aspect of health and disease at individual and population levels. However, our understanding of how it had evolved is deeply flawed. The scientists studying the evolution of aging are divided into two seemingly irreconcilable groups, and current evolutionary theories of aging cannot even consistently answer basic questions such as why most species age, but some do not. This thesis provides a critical overview of current evolutionary theories of aging from both sides of this deeply divided field and attempts to offer an alternative view free from the problems of contemporary theories. The practical part presents three different agent-based models, each providing novel insights into how aging evolved, and analyzes human mortality data comparing male and female aging rates. Overall, this thesis presents five core findings. First, assuming that non-aging phenotype requires coordinated interaction of several fine-tuned molecular pathways and aging does not, aging individuals can easily outcompete non-aging individuals in a sexually reproducing population provide there is an assortative mating with respect to non-aging/aging phenotype. Second, the evolutionary benefits of non-aging are much greater for organisms with simultaneous hermaphroditism than for species with genuine males and females. Third, if some aging trade-offs, such as reproductive trade-off, exist, they are the main factors deciding the course of evolution of aging. Fourth, for a given species, there is likely an optimal length of lifespan mitigating genetic drift. Fifth, even though males have higher mortality rates across all age-groups than females, the male and female rates of aging are approximately the same. Taken together, these results provide a new direction and opportunities for the research of mechanisms affecting the evolution of specific rates of aging.
