Genetic diversity of fairy shrimp Branchipus schaefferi in the Czech republic

Abstract:

V této práci se zaměřujeme na genetickou diverzitu žábronožky letní v České republice. Cílem bylo nastudovat známé informace o genetické diverzitě velkých lupenonožců a doplnit některé chybějící informace o diverzitě žábronožky letní v ČR. V rámci praktické části práce jsme se tedy pokusili objasnit otázku příbuznosti populací v ČR mezi sebou i v kontextu západní palearktické oblasti. K výzkumu bylo využito celkově 37 vzorků zahrnujících jedince z 19 různých lokalit.¨ Pro molekulární výzkumy byl využit jaderný marker ITS1, a ze získaných sekvencí jsme vytvořili fylogenetické stromy. Analyzované jedince z našeho území se nám úspěšně podařilo dosadit do jedné ze dvou skupin z rozsáhlého evropského výzkumu kolektivu Lukić et al. (2019). Do této skupiny patří naše populace společně s populacemi z Polska, Srbska, Rakouska, Maďarska, Chorvatska a Francie. Tato skupina zároveň koreluje s kladem B pro COI, který pravděpodobně zahrnuje populace pocházející ze stejného glaciálního refugia. Při porovnání českých populací mezi sebou nebyly zjištěny žádné výrazné genetické rozdíly, na základě markeru ITS1 byly zkoumané populace mezi sebou geneticky velmi blízké. Mezi jedinci z vojenských a polních lokalit nebyly zaznamenány žádné patrné odlišnosti. Pro získání podrobnějších výsledků je však zapotřebí rozsáhlejšího výzkumu.

Abstract:

In this thesis we focus on the genetic diversity of fairy shrimp Branchipus schaefferi in the Czech Republic. The aim was to study known information about the genetic diversity of large branchiopod crustaceans and fill in some of the missing information about the diversity of Branchipus schaefferi in the Czech republic. Within the practical part of this thesis we tried to clarify the relationships of Branchipus schaefferi in Czech populations and also in the wider context of the Palearctic area. In this research a total number of 37 samples were used, including fairy shrimps from 19 different localities. For molecular research a nuclear marker ITS1 was used. Phylogenetic trees were then created from obtained sequences. Analyzed specimens from the Czech Republic were successfully placed in one of the two evolutionary groups known from extensive study by Lukić et al. (2019). Our populations belong to the same group with the populations from Poland, Serbia, Austria, Hungary, Croatia, and France. This group also corresponds with the clade B recognized for COI, which probably clusters populations originating from the same glacial refugia. After comparing the Czech population between each other, no significant genetical differences were found, the populations were genetically very similar, based on marker ITS1. Furthermore, there was not found any specific difference between the individuals from military grounds and field areas. For more detailed results, further research is needed.
