Příprava PCR interních standardů pro detekci a diferenciaci kmenů druhu Mycobacterium avium

Abstract:

Jedním z cíl diplomové práce bylo vyrobit PCR interní standard ke stanovování inzerní sekvence IS1245 druhu M. avium. K tomuto úelu jsem použil druhý exon genu pro bramborovou trehalóza syntázu StTS1. Interní standard který vznikl z tohoto genu, jsem naklonoval do komerního T-vektoru (Invitrogen). Po optimalizaci polymerázové etzové reakce jsem pipravil PCR master mix, do kterého jsem pidal 100 molekul interního standardu. U této smsi jsem stanovil senzitivitu reakce na 10 cílových molekul na jednu polymerázovou etzovou reakci. Výsledky této ásti jsou souasn využívány pro detekci M. avium ve veterinárním klinickém materiálu. Tím bylo dosaženo stanoveného cíle. Dalším z cíl byla výroba T-vektoru uplatnitelného pi TA-klonování PCR produkt. Jako materiál pro jeho výrobu jsem vybral plazmid pUC19. Do nho jsem vložil synteticky vyrobenou kazetu, která v sob obsahovala dv štpící místa pro restrikní endonukleázu XcmI. Po štpení rekombinantního plazmidu (nazval jsem ho pUC19-TA) a jeho peištní jsem získal T-vektor. Pro TA-klonování s tímto vektorem jsem navrhl protokol a otestoval jeho úinnost. Po mnoha testech však nebyla pesn stanovena. Vektor sice prokazoval malou klonovací schopnost, ale pro jeho neovené schopnosti nebude pravdpodobn využíván.

Abstract:

One of goals of this diploma paper was to prepare PCR internal standard for determining the insertion sequence IS1245 of M. avium. I used the second exon of the Solanum tuberosum putative trehalose synthase (StTS1) gene for this purpose. I cloned the internal standard created this way to commercial cloning T-vector (Invitrogen). After optimising the polymerase chain reaction, I prepared the PCR master mix containing 100 molecules of the internal standard. I have established the PCR sensitivity applying this mixture onto 10 target molecules per a reaction. The results of this part of the paper are used for M. avium detection in veterinary clinical materials. Thus, I have achieved the set goal. The next goal was the preparation of the T-vector applicable to TA-cloning of PCR products. I chose the pUC19 plasmid as the source for its production. I inserted a synthetic cassette containing two cleavage sites of the restriction enzyme XcmI into this plasmid. I obtained the T-vector after digestion with XcmI and repurifying of a recombinant plasmid (I called them pUC19-TA). I have suggested a protocol for cloning with this vector and I have tested its efficiency. However, it stays undetermined after many tests. The vector showed small cloning capacity but for its properties that havent been tested it will not probably be used
