Analysis of signal pathways components in single animal cells

Abstract:

Analýza jednotlivých buněk zahrnuje široké spektrum analytických metod, které umožňují charakterizaci jejich chemického složení. Tato práce popisuje několik rozdílných přístupů pro analýzu komponent signálních drah apoptózy v jednotlivých živočišných buňkách. V první části práce bylo vyvinuto miniaturizované zařízení pro detekci kaspázových proteáz v jednotlivých myších buňkách. Metoda využívá bioluminiscenci založenou na Photinus pyralis luciferáze a detekci pomocí fotonásobiče pracujícího v režimu počítání fotonů. Toto zařízení bylo následně použito pro studium neapoptotických funkcí jednotlivých kaspáz během diferenciace osteoblastů MC3T3 E1 buněčné linie. Druhá část se zaměřuje na přípravu katodoluminiskujících nanočástic, které mohou být využity pro specifické značení v korelativní světelné a elektronové mikroskopii. Byly syntetizovány dva typy částic dopovaných lanthanoidy – ceriem dopovaný lutecito-hlinitý granát a europiem dopovaný fluorid yttrito-sodný. Částice byly povrchově modifikovány a konjugovány s protilátkami proti Fas ligandu, klíčovému iniciátoru apoptózy. Vlastnosti konjugovaných částic byly testovány na buňkách a tkáňových řezech. Poslední část pojednává o analýze aktivní migrace rakovinných buněk, která úzce souvisí s jejich metastatickým potenciálem. Bylo vyvinuto pasivní mikrofluidické zařízení pro sledování migrace a deformace buněk v úzkých pórech. Zařízení obsahuje řady pilířků kruhového průřezu, které imitují póry v tkáních. Aktivní migrace buněk je pak stimulována difuzním gradientem nutrientů v migračním kanálku. Zařízení bylo testováno s HeLa buňkami, které byly transfekovány fluorescenčním sensorem buněčného cyklu.

Abstract:

The single-cell analysis comprises a wide spectrum of methods that characterize the chemical composition of individual cells. This thesis describes several approaches for the analysis of components of apoptotic signal pathways in individual animal cells. In the first part, a miniaturized device for the detection of caspase proteases in single murine cells was developed. The device utilizes bioluminescence chemistry based on Photinus pyralis firefly luciferase and photon counting for the detection. This device was then used for studying non-apoptotic functions of caspases in the differentiation of MC3T3 E1 osteoblastic cells. The second part focuses on the preparation of cathodoluminescent nanoparticles that can be used as specific labels in correlative light and electron microscopy. Two types of lanthanide-doped particles were synthesized – cerium doped lutetium aluminum garnet and europium doped sodium yttrium fluoride. The particles were surface-modified and conjugated with antibodies against Fas ligand, one of the key initiators of apoptosis. The properties and specificity were tested on cells and tissue sections. The last part focuses on the analysis of active cancer cell migration which is closely related with metastatic potential. A passive microfluidic device for monitoring of cell migration and deformation in confining spaces was developed. The device comprises circular pillar arrays that mimic the pores in tissues and active cell migration is induced by a diffusion gradient of nutrients. The device was tested with HeLa cells transfected with a fluorescent cell cycle sensor.
