Phytoremediation of nitrosocompounds

Abstract:

Práce vznikla za účelem porovnání citlivosti pěti odrůd lnu setého (Linum usitatissimum) k fytotoxickým účinkům nitrolátek - 2,4,6-trinitrotoluenu (TNT) a nitroglycerinu (NG) pomocí ekotestu založeném na míře inhibice elongace kořínků klíčních rostlin v médiích s různou koncentrací těchto látek. Na základě výsledků z těchto testů byly vytvořeny dvojice grafů, první ukazuje nárůst kořínků, druhý inhibici. Ke zjištění schopnosti degradace nitrosloučenin rostlinami lnu byla z odrůdy Flanders odvozena kalusová kultura, která následně byla převedena na suspenzní buněčnou kulturu. Na té byla testována schopnost degradovat 2,4,6-trinitrotoluen. Viabilita buněk v průběhu degradace byla měřena na základě oxidačních přeměn TTC na červený formazán, intenzita zbarvení roztoku byla měřena spektrofotometricky.Testy ukázaly, že nitroglycerin je ve srovnatelných koncentracích méně toxický než trinitrotoluen, jeho toxicita v přírodě však může být vyšší vzhledem k jeho výrazně vyšší rozpustnosti ve vodných roztocích. Byly určeny odrůdy lnu s nejlepší schopností odbourávat nitrosloučeniny ze zamořených ploch.

Abstract:

The aim of this diploma thesis was to compare the sensitivity of five varieties of cultivated flax (Linum usitatissimum) to the phytotoxic effects of following nitrosocompounds: 2, 4, 6 – trinitrotoluene (TNT) and nitroglycerine (NG). The sensitivity was determined by an ecotest which measured the inhibition of the elongation of germinating plant´s rootlets in fluids with different concentrations of the above mentioned substances. The results of the test were shown in two charts: one depicting the growth of rootlets, the other one the inhibition. The ability of decomposition of nitrosocompounds by the flax plants was tested on a callus culture, which was derived from Flanders variety of flax and subsequently converted to a suspension cellular culture. Viability of cells during the decomposition was measured on the basis of oxidation conversion of TTC to red formazan; the intensity of the solution coloration was determined spectrophotometrically. The tests showed that in similar concentrations, nitroglycerine is much less toxic than trinitrotoluene is. However, the toxicity of nitroglycerine in nature can be higher due to its extremely higher water solubility. The flax species with the best ability to decompose nitrosocompounds from contamined areas were chosen.
