Abstract:

Pro rostliny, jako přisedlé organismy, světlo představuje nejen zdroj energie pro proces fotosyntézy, ale především zdroj informaci o jejich okolí. Denní fluktuace intenzity slunečního záření a změny v jeho spektrálního složení kriticky ovlivňují jejich růst a vývoj rostlin. I přes veškeré nové poznatky ve studiu tohoto zásadního enviromentálního faktoru není signální mechanismus světla na molekulární úrovni v rostlinách zcela pochopen. V této práci s názvem: “Light-responsive proteins and their role in plant development” jsou v první teoretické části shrnuty nejnovější poznatky o centrálním biochemickém oscilátoru rostlin – cirkadiánních hodinách, které synchronizuje vnímaní času rostlinami, jeho roli v regulaci rostlinného metabolismu – především fytohormonu cytokininu, interakce s molekulou peroxidu vodíků. Poslední teoretická kapitola je zaměřena na roli světla v procesu chladové aklimatizace. Role fytohormonu cytokininu v abitotickém stresu, a role peroxidu vodíku jako signální molekuly rostlin byly shrnuty ve dvou přehledových článcích. Jelikož v podmínkách kultivačních komor nelze mimikovat přirozené diurnální oscilace rostlin, v první studii bylo využito mutantních rostlin fotoreceptoru fytochromu modelového organismu Arabidopsis thaliana. Pomocí LC-MS profilování bylo identifikováno 300 proteinů, jejichž role v diurnálních rytmech nebyla doposud v rostlinách standardního typu Arabidopsis thaliana popsána. Zároveň tato analýza zdůraznila regulace cirkadiáního systému v interakci signalizaci fytohormonu cytokininu a peroxidů vodíku. V následující části práce byla sledována role nízké intenzity světla v procesu chladové aklimatizace. Jelikož většina studií zabývajících se chladovou aklimatizací rostlin sleduje pouze roli faktor teploty na tento proces, aplikace kombinace nízké intenzity světla a nízké teploty přiblížila studium tohoto procesu do podmínek, v nichž se rostliny přirozeně nachází. Poslední části této práce zahrnuje studie zabývající se především interakci fytohormonu cytokininu a metabolismu reaktivních forem kyslíku, na nichž se autorka této práce podílela spoluautorstvím.

Abstract:

For plants as sessile organisms, light represents a source of energy for photosynthesis, as well as a source of information about their surroundings. Daily fluctuations of light intensity and changes in its spectral composition critically affect plants' growth and development. Despite all the new findings in the study of this essential environmental factor, the signaling mechanism of light in plants is not fully understood. This work entitled: "Light-responsive proteins and their role in plant development" briefly summarizes the latest knowledge about the central biochemical oscillator of plants - the circadian clock, which synchronizes the perception of time by plants, its role in the regulation of metabolism - especially that of phytohormone cytokinin, and hydrogen peroxide. The last theoretical chapter is focused on the role of light as a signal molecule in the process of cold acclimatization. The results of the thesis are subdivided into three parts that correspond with individual aims and objectives. The first objective follows the role of light in proteome diurnal oscillations. Arabidopsis thaliana mutant lines with the disrupted perception of red light were used to study these diel changes in protein abundance. Using LC-MS profiling, 300 proteins were identified whose role in diurnal rhythms has not yet been described. At the same time, this analysis emphasized the regulation of the circadian system in the interaction of the phytohormone cytokinin and hydrogen peroxide. Next, the role of low-light intensity in the process of cold acclimatization was monitored. The combination of low PPFD and low temperature provided a unique experimental setup to plant´natural environment. The last part of the results outlines miscellaneous publications co-authored by the author in the course of her Ph.D. study. These works share the topic of plant signaling, including response to cytokinin and the role of reactive oxygen species metabolism.
