The prevalence of the HLA antigens at the blood donors group of the Blood and Tissue Department of Faculty Hospital Brno

Abstract:

Bakalářská práce se zabývá zastoupením HLA alel I. třídy v souboru dárců krve TTO FN Brno. HLA systém člověka představuje vysoce polymorfní soubor molekul, sloužící jako specifické antigeny na povrchu buněk. Zajišťuje tkáňovou neslučitelnost mezi jedinci a účastní se také imunitních reakcí. Cílem práce je zmapovat nejčastěji se vyskytující HLA alely, srovnat jejich frekvence výskytu s jinými populacemi a zaměřit se na zastoupení jednotlivých HLA alel ve vztahu k zastoupení skupinových znaků v systému ABO za účelem zajištění úspěšné hemoterapie. Vyšetření HLA znaků bylo provedeno lymfocytotoxickým testem, do testovaného souboru dárců krve bylo zahrnuto 200 jedinců. Výsledkem práce je statistická analýza frekvence výskytu jednotlivých HLA alel, nejčastější alely v testovaném souboru jsou HLA-A2, A1, B7, B44, B35, Cw7. Zastoupení HLA alel se lišilo mezi etnickými populacemi. Ve vztahu HLA alel k systému ABO byly patrné určité odlišnosti, vzhledem k malému počtu testovaných jedinců v rámci dané krevní skupiny však tyto výsledky nemají příliš velký význam. Bakalářská práce přispívá k získání přehledu o HLA systému, o nejčastějších HLA alelách a o rozdílech sledovaných znaků mezi populacemi. Důraz je v této problematice kladem především na zajištění účelné hemoterapie u imunizovaných polytransfundovaných pacientů.

Abstract:

Bachelor thesis deals with the existence of HLA class I alleles in the group of blood donors TTO FN Brno. Human HLA system is highly polymorphic set of molecules serving as the specific antigens on the cell surface. The HLA systems enables tissue incompatibility between individuals and also participates in immune responses. The aim is to map the most frequent HLA alleles to compare their frequency with other populations and focus on the representation of individual HLA alleles in relation to the representation of groups of ABO system in order to ensure a successful haemotherapy. HLA examination was performed by using the lymphocytotoxicity technique, the tested group of blood donors included 200 subjects. The result of the thesis is a statistical analysis of the frequency of the occurrence of the individual HLA alleles, the most frequent allele in the test group are HLA-A2, A1, B7, B44, B35, Cw7. The existence of HLA alleles was different between ethnic populations. In relation of HLA alleles and the ABO system some differences were observed, because of the small number of tested individuals within the specific blood group, however, these results do not seem to be very important. Bachelor thesis contributes to gain an overview of the HLA system, the most frequent HLA alleles and the differences between populations. The focus in this issue is put mainly on ensuring the effective haemotherapy of the immunized polytransfunded patients.
