Functional genomics of Leishmania Virulence

Abstract:

The family Trypanosomatidae unites unifagellate parasitic protists. This group is intensively studied because of their peculiar molecular and biochemical features as well as medical or economic importance of some trypanosomatids. In this thesis, I reviewed molecular characteristics of Crithidia and Leishmania genera and discussed parasite virulence. Five publications with my co-authorship, which I included here describe molecular aspects of biology of these trypanosomatids. Two publications describe new conditional gene expression system in Leishmania. One of them illustrates details of its work. The second paper describes potential limitations that conditional gene expression systems might have in the studies of the development of Leishmania. Three other publications focus on identification of novel virulence factors in Leishmania and mechanisms of thermoresistance in Crithidia. One concerns the nuclear genome of Leptomonas pyrrhocoris, a monoxenous relative of the human parasites of the genus Leishmania; I contributed to the investigation of novel virulence factors. The second paper being a logical continuation of the previous one is focused on characterization of a gene previously identified as potentially important for Leishmania?s infectivity. The last one describes molecular mechanisms of thermal resistance as a pre-adaptation to the dixenous life cycle of the insect trypanosomatid Crithidia thermophila. Genomics, transcriptomics and molecular biology approaches can bring new insights to the biology of parasites and hosts and can be used to identify potential virulence factors in Leishmania.

Abstract:

Čeleď Trypanosomatidae zahrnuje jednobičíkaté parazitické prvoky. Tato skupina je intenzivně studována, neboť vykazuje neobvyklé molekulární a biochemické znaky a někteří její zástupci jsou důležití z medicínského či ekonomického hlediska. Ve své dizertační práci jsem shrnula molekulární charakteristiky rodů Crithidia a Leishmania a virulenci posledně jmenovaného rodu. Pět publikací, které jsem do této práce zahrnula, popisují molekulární aspekty biologie zmíněných trypanosomatidů. Dvě publikace se zabývají novým expresním systémem u rodu Leishmania. Jedna z nich detailně ilustruje, jakým způsobem tento systém funguje, zatímco druhá se zaměřuje na možná omezení, které by tento typ expresních systémů mohl mít při použití u různých životních stádií rodu Leishmania. Další tři publikace se zaměřují na identifikaci nových virulentních faktorů u rodu Leishmania a teplotní rezistence u rodu Crithidia. První popisuje jaderný genom monoxenního trypanosomatida Leptomonas pyrrhocoris, který je příbuzný lidským patogenům rodu Leishmania; já jsem se zde podílela na výzkumu nových virulentních faktorů. Druhá publikace logicky navazuje na tento projekt a je zaměřena na charakterizaci genu dříve identifikovaného jako potenciálně důležitý faktor pro infektivitu rodu Leishmania. Poslední z těchto tří publikací popisuje molekulární mechanismy teplotní rezistence jako preadaptaci při přechodu na dixenní životní cyklus u hymzího trypanosomatida druhu Crithidia thermophila. Genomika, transkriptomika a molekulární biologie jsou mocné přístupy, které mohou přinést nové vhledy do biologie parazitů rodu Leishmania a jejich hostitelů, popřípadě mohou být použity k identifikaci potencionálních virulentních faktorů.
