Molecular and morphological study of odontoblasts and their differentiation from human embryonic stem cells

Abstract:

Odontoblasty jsou buňky tvořící zubní dentin, hlavní mineralizovanou zubní hmotu. Tyto buňky jsou v zubech zachovány i po prořezání zubu během dospělosti. Odontoblasty vznikají během procesu odontogeneze diferenciací z buněk neurální lišty. Tato skutečnost je zapříčiněna složitou interakcí mezi dentálním epitelem a buňkami neurální lišty, které k dentálnímu epitelu v rámci migrace embryem doputovaly. Cílem této práce je jednak detailně popsat strukturu a funkci odontoblastů, ale také diferenciace buněk neurální lišty z lidských embryonálních kmenových buněk a jejich stimulace do odontogenní linie. V rámci jedné publikace této práce jsou prezentovány nové strukturní a funkční atributy odontoblastů a dentinu. Z hlediska morfologie bylo popsáno zúžení odontoblastů "krček" a jeho opětovné rozšíření "odontopod", nacházející se v těsné blízkosti vznikajícího predentinu a vztah těchto struktur k přilehlé síti kapilár a Tomesovu vláknu. U těchto buněk byla zjištěna exprese dvou nových proteinů (TRPM5 a PIEZO2), které se mohou spolupodílet na senzitivitě zubní pulpy. V rámci druhé publikace byly zdokonaleny postupy při diferenciaci buněk neurální lišty z embryonálních kmenových buněk, byla popsána obnova heterogenity exprese receptoru CD271 po jejich purifikaci metodou průtokové cytometrie a byla zjištěna zvýšená exprese genu MSX1, po stimulaci cytokinem BMP4 (charakteristický jev pro ranou odontogenezi). Výsledky objasňují vývojový vztah mezi odontoblasty a strukturou dentinu a popisují nové členění odontoblastů. Zjištění přítomnosti proteinů TRMP5 a PIEZO2 podporuje teorii odontoblastů jako buněk podílejících se citlivosti zubní pulpy. In vitro experimenty byl zdokonalen protokol diferenciace buněk neurální lišty a proveden prvotní experiment pro možnost budoucího získávání odontoblastům-podobných buněk z pluripotentních kmenových buněk.

Abstract:

Odontoblasts are major matrix-producing cells of the tooth. They are preserved inside tooth pulp even after tooth eruption during adult. They are developed by differentiation from neural crest cells during a process called odontogenesis by complicated interaction between dental epithelium and neural crest cells which migrated to this location. The aim of this thesis is to make detailed visualisation of odontoblasts in relation to dentin and to obtain neural crest cells from human embryonic stem cells and stimulate them to differentiate into odontogenic lineage. New structural and functional atributes of odontoblasts are described in this work. From a morphological point of view we have described that the odontoblast body constricts near predentin (neck) to widen again in a place we call odontopode and we have described relation of these novel parts to the net of blood vessels and Tomes' fiber. We have identified expression of two novel proteins (TRPM5 and PIEZO2) in odontoblasts, which might be responsible for pulp sensitivity. In the second publication we have improved the protocol of differentiation of neural crest cells from human embryonic stem cells, we have described regeneration of heterogeneity in expression of CD271 after fluorescence-activated cell sorting and increased expression of MSX1 gene after BMP4 treatment (characteristic phenomenon for early odontogenesis). The results elucidate developmental relation between odontoblasts and dentin structure, describe new odontoblast's compartments and the expression of TRPM5 and PIEZO2 supports the theory of odontoblasts being responsible for pulp sensitivity. By in vitro experiments we have improved the differentiation protocol of neural crest cells and initial experiment of future obtaining of odontoblasts-like cells was performed.
