Application Of Dithionicotinic Acid As a Chromogen For Spectrophotometric Determination Of Cholinesterase Activity

Abstract:

Cílem teoretické části této diplomové práce je vypracování literární rešerše na téma cholinesterázy a jejich inhibitory. V jednotlivých podkapitolách se věnuje charakteristice jejich struktury, funkce a mechanismu katalytického působení cholinesteráz a možnostem ovlivnění jejich aktivity na různých úrovních. V poslední části se věnuje jednotlivým analytickým metodám, kterými lze stanovit aktivitu a inhibici cholinesteráz, z pohledu historie i současnosti. Experimentální část práce se skládá z několika dílčích podcílů. V první části se věnuje zpracování odebrané krve a dalším úpravám před samotným stanovením. Dalším podcílem je testování různých reakčních podmínek za účelem získání optimálních podmínek pro spektrofotometrické měření aktivity cholinesteráz s využitím dithionikotinové kyseliny (DTNA) jako chromogenního činidla. Dále se práce zabývá porovnáním chromogenních činidel DTNA a DTNB a vyhodnocením vhodnosti jejich použití. Součástí práce je také stanovení inhibiční účinnosti vybraných inhibitorů a jejich porovnání se standardními látkami. Posledním podcílem experimentální části práce je stanovení rozdělovacího koeficientu vybraných látek v systému n-oktanol:voda.

Abstract:

The aim of the theoretical part of this thesis is to elaborate a literature review on the subject of cholinesterases and their inhibitors. The individual subchapters deal with the characteristics of its structure, function, and mechanism of the catalytic action of cholinesterases and the possibilities of influencing their activity at different levels. The last part describes individual analytical methods to determine the activity and inhibition of cholinesterases, in terms of history and the present. The experimental part of the thesis consists of several sub-aims. The first part deals with the processing of the collected blood and other adjustments before the determination. Another objective is testing of different reaction conditions for the purpose of obtaining optimal conditions for the spectrophotometric measurement of cholinesterase activity using dithionicotinic acid (DTNA) as a chromogenic agent. Furthermore, the comparison of DTNA and DTNB chromogenic reagents and their suitability for use was tested. Also, the inhibitory activity of selected inhibitors was determined and compared with standard substances. The last part is the determination of the partition coefficient of selected substances in the n-octanol/water system.
